摘  要:目的探讨α-突触核蛋白A53T转基因帕金森病模型小鼠嗅觉及焦虑样行为的改变,以及其与运动的相关性。方法选用背景为C57BL/6的α-突触核蛋白A53T转基因黑色小鼠和C57BL/6的正常野生型黑色小鼠杂交获得的6月龄杂合子A53T转基因小鼠作为实验组,以正常野生型小鼠为对照组,采用高架十字迷宫实验检测两组小鼠开放臂与封闭臂的活动时间、穿梭次数之比及其各自的活动总距离,嗅闻识别能力实验检测两组小鼠嗅闻新旧垫料的时间以及活动的总距离,精细嗅闻识别实验检测两组小鼠对两种不同混合浓度的气味的正确识别率,转棒实验检测两组小鼠停留在棒上的时间。结果与对照组比较,实验组高架十字迷宫实验中小鼠开放臂活动的时间明显较封闭臂长,差异有显著性(t=2.20,P<0.05);嗅闻识别能力实验中小鼠嗅闻新旧垫料的时间、精细嗅闻识别能力实验中小鼠对两种不同混合浓度气味的正确识别率、转棒实验中小鼠停留在转棒上的时间与对照组比较,差异无统计学意义(P>0.05)。结论 A53T转基因小鼠早期出现嗅觉障碍并且早于运动障碍发生。Objective To study the changes in olfactory and anxiety-like behavior and its correlation with movement in A53 Ttransgenic Parkinson's disease model in mice. Methods The 6-month-old experimental mice were generated on a C57BL/6genetic background with wild phenotype,and the wild type mice served as the control group.Using elevated plus-maze test to measure the duration time percentage of open arms and closed arms,travel times and the total distance.Olfactory recognition ability test was used to detect the time that mice smelled the new and old bedding and their total distance.In the fine odor discrimination test,the correct recognition rate of two odors was recorded.The rotarod test was adopted to measure the time that mice staying on the rod. Results Comparing with the control group,the experimental mice spent more time in open arms than close arms in the elevated plus-maze test(t=2.20,P〈0.05);in the olfactory discrimination test,the mice spent similar time on the old and the new bedding(P〉0.05);in fine odor discrimination test,the mice exhibited no significant differences between two odors which were mixed by different concentration(P〉0.05);and in the rotarod test,the time that the experimental mice stayed on the rotarod had no significant changes(P〉0.05). Conclusion A53 Ttransgenic mice exhibit olfactory deficits in early phase,being prior to movement disorders.
