摘  要:目的探讨转录因子叉头框蛋白O1(FOXO1)对人宫颈癌细胞增殖能力的影响。方法选取宫颈癌组织样本108例和正常宫颈组织76例。采用免疫组化方法检测FOXO1在宫颈癌组织中的表达,免疫印迹方法检测转染过表达质粒或干扰质粒后不同标志物的表达变化,流式细胞术检测FOXO1对宫颈癌细胞增殖能力的影响。分析FOXO1表达与宫颈癌临床病理分期的关系。结果FOXO1阳性表达率在宫颈癌组织高于正常宫颈组织(75.9%vs.1.3%)(P<0.01)。FOXO1的表达与肿瘤分期密切相关(P<0.05)。免疫印迹显示,敲低FOXO1的表达后导致细胞周期的促进蛋白CDC25B、周期素B1和周期素D1表达下调,而细胞周期抑制蛋白p21^(Cip1)和p27^(Kip1)表达增加。而过表达FOXO1显著地提高宫颈癌细胞的增殖能力。结论 FOXO1在宫颈癌中表达增高;其表达增高及降低分别可产生促进宫颈癌细胞增殖及抑制细胞增殖的作用。Objective To investigate the expression and significance of, transcription factor forkhead box protein O1 (FOXO1) in cervical cancer. Methods The expression of FOXO1 was detected by immunohistochemical analysis in cervical cancer of 108 patients (group A) and normal cervical tissues of 76 cases(group B). The difference between the overexpression transfected plasmids and the interference plasmids in cervical cancers was analyzed by Western blot. Cell viability was measured using cell proliferation kit Ⅱ (XTT) and cell cycle was determined by fluorescence-activated cell analysis. The relationship of FOXO1 expression and clinicopathological parameters was analyzed, Results Th positive expression rate of FOXO1 was higher in group A than that in group B(75.9% vs. 1.3%)(P〈0. 01), which was closely related with clinicopathological staging of cervical cancer (P〈0. 05). The depletion of FOXO1 by RNA interference in FOXOl-overexpressed cervical cancer cells caused significant inhibition of cell proliferation. This inhibitory phenomenon was associated with the reduced expressions of cyclin B1, cyclin D1 and CDC25B, but increased the expressions of p27Kip1 and p21cipl. The overexpression of FOXO1 remarkbly increased the cell proliferation. Conclusion FOXO1 is overexpressed in cervical cancer cells. The overexpression and deleption of FOXO1 in cervical cancer ceils can respectively enhance and inhibit cell proliferation.
