摘  要:目的探讨基质细胞衍生因子-1(SDF-1)对髓核细胞基质金属蛋白酶-13(MMP-13)表达的调控机制。方法体外培养大鼠髓核细胞,应用SDF-1进行干预,以CXC趋化因子受体4、7(CXCR4、CXCR7)中和抗体阻抑SDF-1/CXCR4、7轴,检测MMP-13和活性的改变,荧光素酶报告基因检测法验证SDF-1与MMP-13作用关系。结果SDF-1促进髓核细胞MMP-13表达,CXCR4中和抗体抑制MMP-13的表达和活性改变(P〈0.05)。SDF-1增加MMP-13启动子的活性2.7倍,CXCR4抑制剂可降低MMP-13启动子活性3.8倍(P〈0.05)。结论SDF-1结合CXCR4后,激活MMP-13启动子并上调其表达水平。Objective To investigate the mechanism of stromal cell derived factor - 1 ( SDF - 1 ) regulating the expression of matrix metalloproteinase - 13 ( MMP - 13 ). Methods Rat nucleus pulposus (NP) cells were isolated and co - cuhured with SDF - 1. CXC chemokine receptor - 4 ( CXCR4 ) and CXCR7 - neutralizing antibody were incubated respectively to inhibit the SDF - 1/CXCR4 and SDF - 1/ CXCR7 axis. The expression of MMP - 13 and activity was detected. Lueiferase activity was measured with the Lueiferase reporter assay system to investigate the correlation between SDF - 1 and MMP - 13. Results SDF- 1 induced the expression of MMP - 13, while the expression of MMP - 13 could be blocked by CXCR4 neutralizing antibody ( P 〈 0. 05 ). SDF - 1 increased the lueiferase activities of MMP - 13 promot- er by 6. 2 - fold. Addition of CXCR4 inhibitor reduced the lueiferase activities by 6. 2 - fold ( P 〈 0.05 ). Conclusion SDF - 1 acted through CXCR4 to increase MMP - 13 promoter activity and upregulate MMP - 13 expression.
