摘  要:目的探讨皮质骨和松质骨Wnt/β-连环蛋白(B—catenin)信号通路及相关分子表达的差异。方法通过小鼠在体和离体实验,观察皮质骨和松质骨Wnt/β-catenin通路及相关因子B—catenin、核心结合蛋白因子2(Runx2)、成骨相关转录因子抗体(Osterix)等表达水平的差异。结果免疫组织化学结果显示小鼠皮质骨B—catenin、Runx2、Osterix阳性表达量均明显低于松质骨l(20.30±1.21)%比(25.80±1.10)%、(10.20±3.05)%比(19.51±2.03)%、(13.90±2.06)%比(23.74±3.33)%,P〈0.05]。细胞免疫荧光和实时定量聚合酶链反应(Real—time PCR)的结果与免疫组织化学结果一致。结论皮质骨的Wnt/β-catenin信号通路及相关分子表达明显低于松质骨。Objective To investigate the difference of Wnt/β- catenin pathway and associating factorsexpression in cortical and trabecularbone. Methods β- catenin and associating factors such as related transcription factor - 2 ( Runx2), Osterixin in cortical and trabecular bone were examined in vitro and in vivo. Results Immunohistochemical staining on micerevealed that the expression levels of 13 - catenin, Runx2 and Osterix in cortical bone were significantly lower than those in trabecular bone [ ( 20. 30 ± 1.21)% vs. (25.80±1.10)%, (10.20±3.05)% vs. (19.51 ±2.03)%, (13.90±2.06)% vs. (23.74 ±3.33)% ; P 〈0. 051. The results of cellular immune fluorescence and real - time quantitative polymerase chain reaction (Real -time PCR) were identical with the Immunohistochemieal staining. Conclusion The activity of Wnt/β - catcnin signaling pathway and associating factors expressionin cortical bone were significantly lower than those in trabecular bone.
