摘 要:目的观察KISS-1在早期结肠癌原发病灶和转移淋巴结中的表达,探讨KISS-1在结肠癌淋巴结微转移中的作用。方法收集我院经手术切除且病理检查证实的65例结肠癌病例,记录患者性别、年龄、吸烟饮酒史、肿瘤性质、浸润程度、淋巴结微转移及Dukes分期等各项临床相关资料。将切除的肿瘤标本和清扫的淋巴结标本行免疫组织化学染色,并提取肿瘤和淋巴结组织的DNA行聚合酶链反应(PCR)扩增和测序,观察KISS—1在结肠癌原发病灶和转移淋巴结中的表达情况,并分析各临床病理特征和患者术后总体生存的关系。结果KISS-1mRNA在65例结肠癌病例的原发灶均表达阳性(100.00%),327站淋巴结中49站(14.98%)表达阳性,196站肠系膜淋巴结中24站(12.24%)表达阳性。影响术后生存和无病生存率的因素包括:Dukes分期(P〈0.01)、肠系膜淋巴结微转移(P〈0.01)、清扫淋巴结总站数(P〈0.01)、清扫肠系膜淋巴结站数(P〈0.05)、病理分级(P〈0.05)。结论检测结肠癌原发肿瘤病灶KISS-1mRNA蛋白表达有助于预测早期结肠癌患者的预后。Objective To investigate the expression of KISS - 1 in primary colorectal cancer and metastatic lymph nodes, and to explore the role of KISS - 1 in the lymph node metastasis of colon cancer. Methods Collected in our hospital from March 2012 to March 2015, menstrual surgical resection and pathology of 65 cases of colon cancer cases were recorded each patient' s sex, age, smoking, drinking histo- ry, the nature of the tumor, invasibn degree, lymph node micrometastasis and dukes staging and the clinical data. The resection of the tumor specimens and lymph node specimens, immunohistochemical staining and extract tumor and lymph node tissue DNA for polymerase chain reaction (PCR) amplification and sequencing, observe KiSS- 1 in colon cancer primary lesion and metastatic lymph nodes in the expression, and analyze the relationship between the clinical pathological factors and postoperative overall survivaI. Results ( 1 ) KISS - 1 mRNA in 65 cases of lung cancer cases of primary tumors were positive ( 100. 00% ) , 327 station lymph nodes in 49 stations ( 14. 98% ) positive, 196 stations in the mesenteric lymph node 24 (12. 24% ) positive expression. (2)The influence of survival and disease -free survival rate of factors including Dukes stage ( P 〈 0. 01 ), mesenteric lymph node mierometastases ( P 〈 0.01 ), lymph node station number (P 〈 0. 01 ) cleaning, cleaning of mesenterie lymph node stations numbers (P 〈 0. 05 ) , pathological grade ( P 〈 0.05 ). Conclusion The detection of colon cancer primary tumor lesions expressed KISS - 1 mRNA protein, and to study its lymph node micrometastasis and prognosis. Can more accurately predict the prognosis of patients with colon cancer, reduce the blindness of postoperative treatment, improve the curative effect.
