摘  要:背景:癫痫与细胞凋亡之间密切相关,癫痫的发生会导致海马神经元出现丢失现象和凋亡,引发一系列程序性细胞死亡的出现。目的:观察骨髓基质干细胞移植治疗癫痫对细胞凋亡产生的影响。方法:45只SD大鼠制备癫痫模型,随机分为3组,即癫痫未移植组、生理盐水组和骨髓基质干细胞组,分别实施未移植治疗和生理盐水注射治疗以及骨髓基质干细胞移植治疗。造模后1,2,4周,免疫组织化学法检测3组动物脑组织Bax阳性细胞数、Bcl-2阳性细胞数及Bax/Bcl-2情况。结果与结论:(1)不同时间生理盐水组Bax、Bcl-2及Bax/Bcl-2值均未出现明显的改变;(2)造模后1,2,4周,骨髓基质干细胞组与癫痫未移植组和生理盐水组比较,骨髓基质干细胞组Bax阳性细胞数、Bax/Bcl-2值均出现显著的下降,Bcl-2阳性细胞数则出现显著的增加(均P<0.05),骨髓基质干细胞组造模后不同时间组内比较,差异有显著性意义(均P<0.05);(3)结果表明,骨髓基质干细胞移植能下调癫痫大鼠脑组织促凋亡因子Bax阳性细胞数,上调抗凋亡因子Bcl-2阳性细胞数,提示其对癫痫所导致的细胞凋亡产生一定的影响,可能减少癫痫所致细胞凋亡的出现。BACKGROUND:There is a close relationship between epilepsy and apoptosis. The appearance of epilepsy can lead to the loss of neurons in the hippocampus, triggering a series of programmed cel death. OBJECTIVE:To investigate the effect of bone marrow stromal stem cel transplantation on apoptosis in epilepsy. METHODS:After modeled to be of epilepsy 45, Sprague-Dawley model rats were randomly divided into three groups, fol owed by given no intervention (moldel group), normal saline (normal saline group) or bone marrow stromal stem cel transplantation (transplantation group). At 1, 2 and 4 weeks after modeling, the number of Bax-positive cel s, Bcl-2-positive cel s and Bax/Bcl-2 were detected by immunohistochemistry. RESULTS AND CONCLUSION:The number of Bax-positive cel s, Bcl-2-positive cel s and Bax/Bcl-2 presented no obvious changes in the normal saline group at different time points. However, the number of Bax-positive cel s and Bax/Bcl-2 in the transplantation group was significantly decreased, while the number of Bcl-2-positive cel s significantly increased compared with the other two groups at 1, 2 and 4 weeks after modeling (P〈0.05). Moreover, the above indicators varied significantly in the transplantation group at different time points after modeling (P〈0.05). These results show that bone marrow stromal stem cel transplantation can affect the apoptosis and effectively reduce the apoptosis in rats with epilepsy by up-regulating the number of Bax-positive cel s and down-regulating the number of Bcl-2-positive cel s.
