摘  要:采用高效液相色谱-二极管阵列检测(HPLC-DAD)法检测亚麻木酚素(SDG)含量时发现,木酚素检测的峰面积相对标准偏差在9.7%左右。为了提高木酚素检测的精密度,运用逐项排除法对影响木酚素检测精密度的因素进行了分析。结果表明,影响木酚素检测精密度的主要因素有碱水解后的蒸馏方式、甲醇溶解样品时的挂壁现象、针式过滤器的使用方法等;并针对性提出了相应的改进措施,使木酚素检测的峰面积相对标准偏差由9.7%左右降到0.95%左右。The experiment of content detection of flaxseed lignans using high performance liquid chromatograph HPLC- DAD found that the peak area relative standard deviation of lignans detection reached to about 9.7%. To improve the precision degree of lignans detection, the itemized exclusive method was applied to analyze influencing factors of the flaxseed lignans detection precision. The result showed that, the main factors which affected the detection precision of flaxseed lignans were: the distillation method after basic hydrolysis,hanging wall phenomenon of methanol dissolved samples, the application method of pin- type filter, etc. The corresponding improvement measures were proposed aimed at the influencing factors above, which reduced the peak area relative standard deviation of flaxseed lignans content detection from 9.7% to 0.95%.
