摘 要:目的研究槐耳浸膏对胃癌耐药细胞(SGC7901/Fu)耐药性的逆转作用及对上皮间质转化(EMT)的影响。方法CCK-8法计算SGC7901/Fu细胞的耐药性;光镜观察细胞形态的改变;realtimeRT-PCR检测EMT相关标记物、转录因子的表达;Western.blot法检测snail.及Zeb2蛋白的表达;激光共聚焦显微镜观察E—cadherin表达及分布。结果0.1-0.5mg/m1浓度范围内,槐耳浸膏能增加sGc-7901/Fu细胞对5-Fu、DDP的敏感性;槐耳浸膏作用后sGc7901/Fu呈上皮样细胞形态改变,伴上皮表型标记物表达下调、间质表型标记物表达上调,侵袭能力增强。结论槐耳浸膏能部分逆转SGC.7901/Fu细胞的耐药性,其机制可能是通过调控EMT实现的。Objective To investigate the huaier's role in reversing chemoresistance and epithelial-mesenchymal transition ( EMT ) of chemo- resistant gastric cancer cells ( SGC7901/Fu ) . Methods CCK-8 method was used to determinate the reversal role of Huaier on the chemoresistance of SGC7901/Fu cells; the realtime RT-PCR was used to determine the expression level of EMT markers and EMT-related transcriptional factors; Western-blot was used to determine the protein level of snail and Zeb2; Confocal microscopy analysis was used to determine the change of expression and distribution of E-cadherin; Transwell analysis was used to the change of invasion ability. Results The combined used of Huaier, at 0.1 - 0.5 mg/ ml concentration, and DDP or 5-Fu could increase the chemo-sensitivity of SGC7901/Fu cells to DDP and 5-Fu; The use of Huaier on SGC7901/ Fu cells resulted in mesenchymal appearance, decreased expression of epithelial markers and increased expression of mesenchymal markers. Conclusion Huaier could partly reverse the chemoresistance of SGC7901/Fu cells, the underline mechanism might include regulating EMT process.
