摘 要:目的通过检测IL-6与骨密度及骨小梁形成之间的关系,进一步验证JAK2/STAT3信号通路在骨质疏松中的作用机制。方法选取100例骨质疏松患者(OP组)和100例非骨质疏松者(NOP组),通过骨密度分析仪测定骨密度,静脉取外周血通过酶联免疫分析方法和全自动生化分析仪检测血清IL-6及碱性磷酸酶(Alkaline Phosphatase,ALP)水平。通过卵巢切除术构建大鼠骨质疏松模型,随机分为未阻断组、JAK/STAT阻断组、假手术组,分离静脉血及处死后分离股骨及胸椎骨通过ELISA及组织HE染色检测IL-6及骨小梁的情况。结果 OP组血清IL-6水平[(193.45±35.78)pg/ml]显著高于NOP组[(114.23±41.12)pg/ml](t=12.183,P<0.001)。大鼠模型JAK/STAT未阻断组骨密度[(0.499±0.21)g/cm2]与ALP水平[(66.43±9.18)U/L]较假手术组均明显降低(F=0.207,22.689;P<0.001),说明造模成功。而大鼠模型JAK/STAT阻断组的骨密度[(0.707±0.19)g/cm2]与ALP水平[(89.67±8.98)U/L]明显高于未阻断组(F=0.215,26.464;P<0.001);HE染色显示JAK/STAT阻断组骨小梁的密度及质量明显高于未阻断组。结论 JAK2/STAT3信号途径影响骨小梁形成,并以此下调骨密度从而在骨质疏松症的发生及发展过程中发挥着重要的作用。Objective This work was to investigate the correlation between IL-6 level and bone trabecula and explore the mechanism of osteoporosis formation through JAK2/STAT3. Methods The data of the osteoporosis patients (OP group) and non-osteoporosis patients (NOP group) were collected and the bone mineral density was detected using dual-energy X-ray absorptiometry. Serum levels of IL-6 and ALP were detected in experiment group and control group by ELISA and biochemical method. Ovariectomized female SD rats as the model of osteoporosis and osteoporotie fracture were divided into three groups: non-blocking group, JAK2/STAT3 blocking group and sham operation group. The IL-6 levels and formation of bone trabecula were detected by ELISA and HE. Results The IL-6 levels were higher in OP group (193.45±35.78 pg/ml) than in NOP group (114.23±41.12 pg/ml) (t = 12.183,P 〈 0.001 ). In osteoporosis model groups, both the BMD (0.499±0.21 g/cm2) and ALP (66.43±9.18 U/L) levels were lower in non-blocking group than that in the sham operation group (F = 0.207, 22.689; P〈0.001). The BMD (0.707±0.19 g/era2) and ALP (89.67±8.98 U/L) levels were obviously higher in JAK2/STAT3 blocking group than that in non-blocking group (F= 0.215, 26.464; P 〈 0.001). The formation of bone trabecula was obviously better in JAK2/STAT3 blocking group compared with non-blocking group. Conclusion The formation of bone trabecula was regulated by JAK2/STAT3 signal pathway, and its function of decreasing the bone mineral density played an important role in osteoporosis.
