摘  要:为探究microRNA-139(miR-139)在体内对小鼠失神经腓肠肌是否具有一定的作用,本研究建立了小鼠腓肠肌失神经萎缩模型,然后将10 nmol miR-139 inhibitors注射至小鼠失神经腓肠肌组织中,14 d后qRT-PCR检测miR-139的表达量,比较肌肉湿重和肌纤维直径,RT-PCR和Western blot检测MHC的表达量,研究miR-139表达改变对肌肉发育的影响。结果显示:与对照相比,miR-139 inhibitors注射后显著增加失神经肌肉萎缩部位的肌纤维直径,说明miR-139对体内肌肉的发育分化过程具有一定的调控作用。To investigate the effects of miR-139 on myofibers of denervated muscle atrophy in mouse, anamyotrophia mouse model of sciatic nerve resection was established. 10 nmol miR-139 inhibitors were injected into the denervated gastrocnemius. 14 days after injection, the expression of miR-139 was detected by qRT-PCR, the wet weight and the cross section diameter of the muscle fibers were compared, and the expression of myosin heavy chain(MHC)was examined by RT-PCR and Western blot analysis between the atrophying skeletal muscle and control. The results showed that the cross section diameter of the muscle fibers in treated group was significantly higher than that of control group. The results suggested that miR-139 may play an important role in muscle growth and differentiation in vivo.
