摘  要:目的:研究天麻川芎(TC)对自发高血压大鼠肾脏损伤及炎症因子的影响。方法:自发高血压大鼠(SHR)分为SHR组、卡托普利组(90 mg·kg^(-1)·d^(-1))、TC大剂量组(18.4 g·kg^(-1)·d^(-1))和小剂量组(9.2 g·kg^(-1)·d^(-1)),WKY为正常对照。灌胃给药8周。给药结束后,化学比色法测24 h尿蛋白、免疫比浊法测尿微量白蛋白的含量;ELISA法测定血浆CRP水平;RT-PCR法测定肾脏TNF-α、IL-6、IL-1βmRNA表达水平;取肾脏做病理切片观察。结果:CT小、大剂量组均可显著降低SHR的24 h尿微量白蛋白水平,CT大剂量组可显著降低SHR的24 h尿蛋白水平;CT小、大剂量组均可显著降低SHR组的血浆CRP水平(P<0.05)及肾脏中的TNF-α、IL-6、IL-1βmRNA的表达水平,P值均小于0.05。肾脏病理切片显示CT使肾小球纤维化明显降低,炎细胞减少,肾小管纤维化减轻。结论:天麻川芎对自发高血压大鼠具有改善肾功能、减轻肾脏损伤的作用,其机制可能与降低CRP及炎症因子水平有关。Objective: To investigate the effects of Tianma and Chuanxiong on kidneys and inflammation factors in spontaneous hypertension rats( SHR). Methods: SHR rats were divided into 4 groups: SHR control,captopril( 90 g·kg^-1·d^-1,large dose( 18. 4 g·kg^-1·d^-1 and small dose( 9. 2 g·kg^-1·d^-1. WKY rats were as normal control group. The medicines were given by i. g. for 8 weeks. The levels of microalbumin( m- ALB) and total protein( U- TP) in 24 h urine were detected by immunoturbidimetry and chemical colorimetry respectively. CRP in plasma was detected by ELISA. The expressions of mRNA of TNF- α,IL- 6 and IL- 1βmRNA in kidney were detected respectively by RT- PCR. Results: The levels of m- ALB and U- TP in 24 h urine were decreased markedly. The decline of the expressions of mRNA of TNF- α,IL- 6 and IL- 1βmRNA in kidneys of large and small CT group was significant and had histologic improvement compared with SHR group. Conclusion: Tianma and Chuanxiong can improve the kidney function,decrease the levels of CRP in serum and inflammatory factors mRNA in kidneys and reduce the degree of hypertensive renal damage.
