摘 要:为了明确海洋细菌GM-1与变异菌株GM-1-2菌株的抗菌特性,以白色念珠菌(Candidaalbicans)为指示菌,采用牛津杯法测定海洋细菌GM-1及其变异菌株GM-1-2菌株的抑菌作用及其稳定性,比较2个菌株发酵液的抑菌作用。结果表明:GM-1和GM-1-2菌株及无菌发酵液对白色念珠菌均具有明显的抑制作用,变异后的GM-1-2菌株抑菌作用下降;两个菌株的抑菌作用在不同条件下稳定性不同。GM-1菌株发酵液100℃处理2h抑菌作用无明显变化,121℃处理20min抑菌作用明显下降;GM-1-2菌株在80℃处理2h抑菌作用无明显变化,100℃处理2h抑菌作用降低,121℃处理2h抑菌作用消失;2个菌株的发酵液的抑菌作用在pH6-8条件与未处理的对照无显著差异,pH高于8或低于6的条件下处理24h抑菌作用显著下降;发酵液经紫外线、光照以及胃蛋白酶、胰蛋白酶和蛋白酶K分别处理不同时间,其抑菌作用与对照无显著差异。表明海洋细菌GM-1及其变异菌株GM-1-2无菌发酵液对白色念珠菌的抑制作用较稳定,菌株变异后抑菌作用及其对温度的稳定性有所下降。In order to define the antibacterial properties of Marine bacteria GM-1 and its mutant strain GM-1-2, using Candida albi- carts as the indicator, the antibacterial activities and the stability of two strains were determind by adopting Oxford cup method. At the same time,the antibacterial activities of fermentation filtrate of two strains GM-1 and GM-1-2 were also compared. Results showed that both two strains and their sterile fermentation filtrate obviously inhibited the growth of Candida albicans, and the inhibitory effect of mutant strain GM-1-2 declined obviously. The stability of two strains varied under different treatment conditions. For example when the sterile fermentation filtrate of GM-1 was heated for 2 h at 100 ℃ ,the antibacterial activity had no remarkable changes,and when the fermentation filtrate was heated for 20 min at 121 ℃ ,its bacteriostatic action reduced remarkably but could be detected. However when the sterile fermentation filtrate of GM-1-2 was processed at 80 ℃ for 2 h, the antibacterial activity had no significant changes, and when it was processed at 100 ℃ for 2 h ,the antibacterial activity was lower,while it disappeared under the condition of 121 ℃ for 20 min. When the fermentation filtrate of two strains were stored under the condition of pH value 6 ~ 8, their bacteriostatic actions had no significant differences in contrast to their own untreated fermentation filtrate, and under the condition of pH value either less than 6 or over 8, their bacteriostatic actions both decreased significantly. In addition, after being treated with ultraviolet radiation, light,pepsin, trypsin or proteinase K for different time respectively, the bacteriostatic activities had no remarkable differences. Conclusively, inhibitory effects on Candida albicans of the sterile fermentation filtrate of marine bacteria strain GM-1 and its mutant strain GM-1-2 had good stability. Besides, the antibacterial activity and the stability of mutant strain GM-1-2 were lower than its original strain.
