摘  要:采用水提醇沉法提取新疆沙枣多糖,并采用DM-18型大孔树脂对粗沙枣多糖进行分离纯化,确定了最佳的分离条件。通过粗沙枣多糖和纯化后的沙枣多糖对DPPH自由基、羟基自由基(·OH)、超氧阴离子自由基(O2-·)的清除能力进行比较。结果表明:在沙枣多糖样品溶液2.0 mg/m L,上样速率为1.5 BV/h,上样液p H值为7.0,上样量为3.0 BV、洗脱剂乙醇浓度为35%、洗脱剂用量为3.0 BV、洗脱速率为1.0 BV/h时,洗脱剂p H值为8时,DM-18型大孔树脂对沙枣多糖的动态吸附率和解吸率分别达到90.23%和92.37%,粗沙枣多糖的纯度为42.33%,纯化后的多糖纯度为70.56%。粗多糖和纯化后多糖对DPPH·、·OH、O2-·均有清除作用,纯化后多糖的抗氧化活性大于粗多糖。Polysaccharide from Elaeagnus angustifolia in Xinjiang was extracted by water-alcohol precipitation. DM-18 macroporous resins were used to isolate the crude polysaccharide, the best condition was confirmed. Antioxidant activities of the crude and purified polysaccharide were evaluated through their capability of scav-enging DPPH·,·OH,O2-·. The result showed that best conditions as follows:2.0 mg/mL of concentration and 7.0 of pH of the solution of Elaeagnus angustifolia polysaccharide, the sample loading amount of 3.0 BV, 1.5 BV/h of the rate, the eluent concentration of 35%,the total quantity of eluent of 3.0 BV, 8 of pH of the solution and eluent rate of 1.0 BV/h. The rates of absorption and desorption could achieve 90.23%and 92.37%under the conditions above. The purity of the crude polysaccharide and the purified polysaccharide were 42.33%and 70.56%. Both of the polysaccharide crude and purified had significant effect on scavenging DPPH·,·OH,O2-·. Furthermore purified polysaccharide had more activity antioxidant than crude polysaccharide.
