辛伐他汀抑制实验性大鼠炎性破骨活动的研究

摘　　要：目的:探讨局部应用辛伐他汀对炎症性骨破坏的影响及其机制。方法:选取8周龄雄性Wistar大鼠50只,髓腔直接暴露法建立条件对照组模型(n=25),载药组于开髓术后0、3、6、9、12、15、18、21 d局部注射1 g/L辛伐他汀溶液建立辛伐他汀载药组模型(n=25);于术后1、3、7、14、21 d各组分别处死5只动物,取后牙区下颌骨组织包埋切片后,X线测量根尖病损面积,免疫组化检测TNF-α的表达。结果:载药组3、7、14、21 d的根尖病损面积明显低于条件对照组(P<0.05);载药组TNF-α表达量除21 d组外,其余均低于条件对照组(P<0.05)。结论:局部注射低浓度的辛伐他汀能够抑制根尖周炎模型中TNF-α的表达,从而抑制破骨活动。AIM：To investigate the effects of local application of simvastatin on inflammatory periapical bone destruction in rats.METHODS：8-week-old male Wistar rats were used in this study.The rats were anaesthe-tized by periotoneal injection of pentobarbital sodium.After anaesthesia,pulp cavity of mandibular first molars at one side was exposed.The contralateral teeth received no treatment and served as blank controls.After pulp cavity expo-sure,the rats were randomly divided into simvastatin and control groups.Rats in simvastatin group received local injec-tion of simvastatin at mesial-buccal subperiosteum of mandibular first molars on day 0,3,6,9,12,15,18,and 21. The rats were sacrificed on day 1,3,7,14,and 21 post-operation.Mandibles were removed and sectioned for immu-nohistochemical observation of TNF-αexpression.The bone loss was detected by X -ray examination.RESULTS：The area of periapical bone lesions in simvastatin group was significantly lower than that of control group on 3,7,14, and 21 d （P 〈0.05）.TNF-αexpression in simvastatin group was significantly lower than that in control group at all time points except on day 21 （P 〈0.05）.CONCLUSION：Local application of simvastatin can inhibit inflammatory periapical bone loss through down -regulation of TNF-αexpression.
