摘 要:目的:研究淫羊藿素(ICT)对体外培养SD大鼠骨髓基质细胞(rBMSCs)增殖与成骨分化的影响。方法:体外分离培养rBMSCs,传代至第4代作多向分化鉴定。分别以10-9、10-8、10-7、10-6、10-5mol/L ICT刺激rBMSCs后3、6、9 d,分别用cck-8及碱性磷酸酶ALP试剂盒检测rBMSCs的增殖及ALP活性;10-9mol/L ICT处理r BMSCs后21 d作茜素红(AR)染色以判断钙结节的形成。结果:原代培养的r BMSCs贴壁生长、呈梭形,能多向分化;ICT明显抑制了r BMSCs的增殖;但增高其ALP活性、钙结节形成。结论:ICT以剂量依赖方式抑制rBMSCs的增殖但促进其分化和矿化。Objective: To investigate the effects of icaritin( ICT) on the proliferation and osteogenic differentiation of rat bone marrow stromal cells( rBMSCs). Methods: rBMSCs were cultured from the bone marrow of SD rats and identified by multilineage differentiation assays. 3,6 and 9 days after the treatment of rBMSCs of passage 4 by ICT at 10- 9,10- 8,10- 7,10- 6and 10- 5mol / L respectively,the proliferation and differentiation of the cells were examined by cck-8 and alkaline phosphatase( ALP) activity assay kit respectively. The calcium nodule formation was observed by alizarin red( AR) staining 21 days after 10- 9mol / L ICT treatment.Results: Primary rBMSCs showed the typical spindle-like shape with attachment growth. rBMSCs could be induced to osteogenic and adipogenic differentiation. The proliferation of rBMSCs was inhibited but ALP activity was enhanced by ICT. 10- 9mol / L ICT incresed calcium nodule formation. Conclusion: ICT can dose-dependently inhibit the proliferation,but promote the osteogenic differentiation of rBMSCs.
