摘　　要：目的:研究淫羊藿素(ICT)对体外培养SD大鼠骨髓基质细胞(rBMSCs)增殖与成骨分化的影响。方法:体外分离培养rBMSCs,传代至第4代作多向分化鉴定。分别以10-9、10-8、10-7、10-6、10-5mol/L ICT刺激rBMSCs后3、6、9 d,分别用cck-8及碱性磷酸酶ALP试剂盒检测rBMSCs的增殖及ALP活性;10-9mol/L ICT处理r BMSCs后21 d作茜素红(AR)染色以判断钙结节的形成。结果:原代培养的r BMSCs贴壁生长、呈梭形,能多向分化;ICT明显抑制了r BMSCs的增殖;但增高其ALP活性、钙结节形成。结论:ICT以剂量依赖方式抑制rBMSCs的增殖但促进其分化和矿化。Objective： To investigate the effects of icaritin（ ICT） on the proliferation and osteogenic differentiation of rat bone marrow stromal cells（ rBMSCs）. Methods： rBMSCs were cultured from the bone marrow of SD rats and identified by multilineage differentiation assays. 3,6 and 9 days after the treatment of rBMSCs of passage 4 by ICT at 10- 9,10- 8,10- 7,10- 6and 10- 5mol / L respectively,the proliferation and differentiation of the cells were examined by cck-8 and alkaline phosphatase（ ALP） activity assay kit respectively. The calcium nodule formation was observed by alizarin red（ AR） staining 21 days after 10- 9mol / L ICT treatment.Results： Primary rBMSCs showed the typical spindle-like shape with attachment growth. rBMSCs could be induced to osteogenic and adipogenic differentiation. The proliferation of rBMSCs was inhibited but ALP activity was enhanced by ICT. 10- 9mol / L ICT incresed calcium nodule formation. Conclusion： ICT can dose-dependently inhibit the proliferation,but promote the osteogenic differentiation of rBMSCs.
