摘 要:目的:探讨人脑胶质瘤干细胞增殖中NOTCH信号的调控作用。方法:采用免疫磁珠法从人脑胶质瘤组织及胶质瘤细胞株中提取、分离人脑胶质瘤干细胞,对细胞进行体外培养,采用荧光定量PCR、免疫组化等方法检测相关指标,观察人脑胶质瘤干细胞增殖中NOTCH信号通路的调控作用。结果:正常脑组织的NOTCH-1蛋白累积光密度值为(294.9±29.5),低度及高度恶性胶质瘤患者的NOTCH-1蛋白累积光密度值分别为(5046.0±120.9)和(11 026.2±169.1)。低度及高度恶性胶质瘤患者的NOTCH-1蛋白表达水平较正常脑组织患者明显提高,且高度恶性胶质瘤患者的NOTCH-1蛋白表达水平较低度恶性胶质瘤患者明显提高,比较差异均有统计学意义(P<0.05)。在CD133+胶质瘤细胞中,NOTCH-1、HES-1及CBF-1基因的表达水平较CD133-胶质瘤细胞明显提高,比较差异均有统计学意义(P<0.05)。结论:NOTCH-1基因在人脑胶质瘤中呈高水平表达;在人胶质瘤干细胞增殖过程中,NOTCH-1、CBF-1和HES-1等关键基因呈高表达,NOTCH信号通路在人脑胶质瘤干细胞增殖过程中起着重要调控作用,值得深入研究。Objective:To study the NOTCH signal regulating role in human brain glioma stem cell proliferation.Method:Using magnetic beads immune method extracted from the organization for human brain glioma and glioma cell line,separated the brain glioma stem cells,cells for in vitro culture,used the fluorescent quantitative PCR and immunohistochemical method to detect the related indicators,observed the NOTCH signal pathway in the brain glioma stem cells proliferation regulation function.Result:Patients with normal brain tissue protein accumulation NOTCH-1 optical density value was(294.9±29.5),low and highly malignant gliomas NOTCH-1 protein accumulation optical density value was respectively(5046.0±120.9) and(11 026.2±169.1).Low and highly malignant gliomas NOTCH-1 protein expression levels significantly higher than patients with normal brain tissue,and highly malignant gliomas NOTCH-1 protein expression levels was significantly increased than low grade gliomas(P〈0.05).In CD133+glioma cells,NOTCH-1,CBF-1 and HES-1 gene expression level of CD133 glioma cells increased significantly(P〈0.05).Conclusion:The NOTCH-1 gene is the high level expression in human brain gliomas.In the process of human glioma stem cells proliferation,NOTCH,CBF-1 and HES-1 key genes has high expression,such as NOTCH signaling pathways in the brain glioma stem cells proliferation plays an important regulatory role in the process,it is worth of further study.
