腰痛宁胶囊中士的宁在家兔体内的药动学研究

摘　　要：目的:研究腰痛宁胶囊中士的宁在家兔体内的药动学规律,探究腰痛宁胶囊全方配伍的合理性。方法:给各组家兔分别灌胃给予马钱子粉(调制)胶囊、腰痛宁胶囊及腰痛宁胶囊配黄酒,采用HPLC测定家兔体内士的宁的血药浓度,DAS 3.2.7药代动力学软件拟合药动学模型并计算统计矩参数。结果:士的宁在家兔体内的代谢符合三室开放模型,t检验结果表明,腰痛宁胶囊组与马钱子粉(调制)胶囊组的AUC_(0-t)、AUC_(0-∞)、CL、V、Cmax有显著性差异,腰痛宁胶囊配黄酒组与马钱子粉(调制)胶囊组的CL、V、Cmax有显著性差异;腰痛宁胶囊配黄酒组与腰痛宁胶囊组药动学参数虽无显著性差异,但AUC_(0-t)、AUC_(0-∞)、Cmax均大于后者。结论:腰痛宁胶囊配黄酒配方合理,既有利于降低毒性,又可保证疗效。Objective：To study the law of pharmacokinetics of strychnine within Yao Tong Ning capsule in rabbits ,and to explore the rationality of the whole prescription on Yao Tong Ning capsule. Methods ： The rabbitts were administered intragastrically semen strychni powder （modulation） capsule , Yao Tong Ning capsule and Yao Tong Ning capsule with yellow wine respectively ; the strychnine in rabbit plasma was determined by HPLC ,and pharmacokinetic models were fitted by DAS 3.2.7 software as well as moment parameter. Results： The kinetic processe of strychnine was three-compartment open model . T test results showed that ： AUC（0-1）, AUC（0-∞） , CL , V and Cmax of Yao Tong Ning capsule had significant differences with semen strychni powder （modulation） capsule ; CL , V and Cmax of Yao Tong Ning capsule with yellow wine had significant differences with semen strychni powder （mod- ulation） capsule also ; AUC（0-1 ） , AUC（0-∞） and Cmax of Yao Tong Ning capsule with yellow wine were greater than Yao Tong Ning capsule though they had no significant difference with each other. Conclusion ： It is reasonable that Yao Tong Ning capsule matches with yellow wine ,which both reduces toxicity and guarantees the effect.
