牛病毒性腹泻病毒的分离与鉴定

摘　　要：为了对牛病毒性腹泻病毒进行分离鉴定,本试验通过对黑龙江省完达山牛场疑似感染牛病毒性腹泻病毒患牛的直肠粪便以及鼻拭子进行无菌采集,样品经常规处理后接种于MDBK细胞,盲传3次以及PCR扩增鉴定,最后对收集到的病毒进行蚀斑纯化试验。结果表明,病毒MDBK细胞出现明显病变,PCR鉴定与预期结果相符,3次蚀斑纯化后分离到该病毒。结论可成功分离到牛病毒性腹泻病毒。With suspected bovine viral diarrhea virus disease ~aterial separation and identifica- tion. In this study, cows of Heilongjiang Province Wanda cattle infected with bovine viral diar- rhea virus were diseased acquisition. Specimens collected were inoculated by MDBK cells and repro- duced blindly 3 generations. The viral RNA was amplified by PCR identification, Finally, the col- lected virus by plaque purification experiments purified virus, Result： The virus inoculation ob- vious lesions on MDBK cell. PCR amplified the expected amplification segment. The virus was iso- lated by 3 times plaque purified. Conclusion： Successfully isolated the bovine viral diarrhea vi- rus.
