摘  要:以多年生武威薄皮核桃茎尖为试材,研究了培养基成分、不同种类和质量浓度的褐变抑制剂、生长调节剂以及转瓶周期对其褐变的影响。结果表明:培养基成分、褐变抑制剂、生长调节剂和转瓶周期对外植体褐变的影响均不同;为抑制褐变及保证茎尖正常分化生长,先将茎尖接种于1/2 MS+2 g/L活性炭的培养基中,置于5℃冰箱中暗培养7 d后取出,再转接于DKW+6-BA(1.0 mg/L)+IAA(0.01 mg/L)+5 m L 20%硫代硫酸钠的培养基中,置于25℃室内光照环境下培养,每隔3-5 d转换1次培养基,能有效抑制褐变。Taking the perennial walnut shoot tips as materials,we have studied the effects of medium ingredients,browning inhibitors of different kinds and concentrations,growth regulators and the rotating period on the browning of explants. The results showed that the effects of ingredients,browning inhibitors and rotating period on the browning of walnuts explants are different. Moreover,we first stem tip into 1 /2 MS + 2 g / L of active carbon medium,which can be placed on the refrigerator during 7 days under 5℃ darkness in order to browning inhibition system and guarantee the growth and normal differentiation of stem tip. Thus,it was inoculated into DKW + 6-BA( 1. 0 mg / L) + IAA( 0. 01 mg / L) + 5 m L 20% Na_2S_2O_3 medium,converted one medium per every 3-5days,which can effectively inhibit browning of walnut.
