摘 要:目的观察母子分离后慢性不可预计性应激诱导的青少期大鼠抑郁样行为学改变、下丘脑-垂体-肾上腺(HPA)轴的变化以及调肝方药加味四逆散(JWSNS)对其的干预作用。方法将24只新生Wistar大鼠(雌雄各半)随机分为空白组、模型组、JWSNS组(16.9 g·kg^(-1))。采用母子分离(PND1~21)后予21 d慢性不可预计性应激刺激的方法造模,以JWSNS进行干预,检测各组大鼠旷场实验中行为学改变、血清促肾上腺皮质激素(ACTH)、皮质醇(CORT)及下丘脑促肾上腺皮质激素释放激素(CRH)浓度变化。结果与空白对照组比较,模型组大鼠体质量、糖水偏爱度均降低(P<0.01),旷场试验中大鼠活跃度、总路程均减少(P<0.01),中央区路程明显减少(P<0.05);血清ACTH、CORT浓度升高(P<0.01,P<0.05)。与模型组比较,JWSNS组大鼠糖水偏爱度显著增加(P<0.01),旷场活跃度显著增加(P<0.01),血清ACTH浓度明显降低(P<0.05),CORT及CRH浓度降低但差异无统计学意义(P>0.05)。结论母子分离后慢性不可预计性应激可诱导青少期大鼠抑郁样行为,加味四逆散可改善青少期大鼠抑郁样行为并在一定程度上调节HPA轴功能,具有一定抗青少期抑郁症的作用。Objective To observe the effect of maternal deprivation following with chronic unpredictable stress on the behavior and the hypothalamic-pituitary-adrenal(HPA) axis in adolescent rats,and to investiage the intervention of modified Sini San(MSS). Methods A total of 24 newborn Wistar rats(male in half) were randomly divided into control group,model group,and MSS group(16.9 g·kg^(-1)). After separated from mother from postnatal day 1 to 21,the rats were given chronic unpredictable stress for 21 days to establish the animal model of depression. During the modeling,the depressed rats received oral use of MSS. We observed the behavior changes in rats by open-field test,and measured the serum concentrations of adrenocorticotropic hormone(ACTH), corticosteroid(CORT), and corticotropin-releasing hormone(CRH) with radioimmunoassay. Results Compared with the blank control group,rat body weight and sucrose preference in the model group were decreased(P 0.01),activity and total distance showed by the open-field test were significant decreased(P 0.01)and the central distance was significantly reduced(P 0.05);serum ACTH and CORT levels were increased significantly(P 0.01,P 0.05). Compared with the model group,the sucrose preference index of MSS group was significantly increased(P 0.01),open-field activity was significantly increased(P 0.01),serum ACTH concentrations were significantly decreased(P 0.05),and CORT and CRH levels were decreased but the differences were not statistically significant(P 0.05). Conclusion The method of maternal deprivation following with chronic unpredictable stress can be applied to induce the depression-like behavior in adolescent rats. To some extent,the modified Sini San can improve depression-like behavior and regulate HPA axis function of adolescent stress rats.
