红化妆景天的组织培养与快速繁殖

摘　　要：景天科植物具有良好的景观观赏效果，是优良的地被植物．本试验以红化妆（Echeveria cv Victor）景天植物的离体叶片作外植体进行组织培养，探究了不同激素配比对红化妆景天愈伤组织诱导、分化、生根等的影响，以期建立快速繁殖体系．研究表明：（1）叶片最适灭菌方式：先用75％酒精对叶片消毒30S，无菌水冲洗，然后用0．1％HgCl2溶液消毒5min，无菌水漂洗4～5次；（2）诱导愈伤组织的基本培养基为MS，激素配比为1．0mg／LNAA、1．5mg／L6-BA，培养40d可获得较多的外植体；（3）不定芽诱导的培养基基本培养基为MS，激素配比为1．0mg／LNAA、3．0mg／L6-BA，培养50d可获得较多的不定芽；（4）试管苗生根诱导的基本培养基为MS，激素配比为3．0mg／L6-BA，且植株生长健壮；（5）试管小苗移到基质中50d后，成活率可达85％．Plants of Crassulaceae as an excellent ground covers have the ornamental value,leaf of Echeveria cv Victor were used for explants in study of organ culture to establish a rapid technique system of pmopagation in vitro.The results were asfollow ： （ 1 ） the optimal sterilization of leaves ： At first, disinfecting the blade 30 s with 75% alcohol, sterile water, then, disinfection 5rain with 0.1% HgCl2 solution, rinsed 4-5 times with sterile water; （2） Basic medium for callus induction was MS,hormone combination as 1.0 mg/L NAA,1.5 mg/L 6- BA can get more explant after 40 day＇s culture; （3）Iniation medium： MS ＋ 1.0 mg/L NAA ＋ 3.0 mg/L 6-BA can get more adventitious buds after 50 day＇s culture; （4）Basic medium for rooting was MS, hormone combination as 3.0 mg/L 6-BA ; （5） T he survival rate of tube seedlings moved matrix can reach up 85%.
