DNMT1与EZH2在急性髓系白血病中的表达及相关性研究

摘　　要：目的：探讨DNA甲基转移酶DNMT1与EZH2在急性髓系白血病（AML）中的表达及其相关性。方法：采用荧光定量PCR技术检测50例AML患者和30例正常供者骨髓细胞中DNMT1与EZH2mRNA的表达水平,分析DNMT1表达水平与临床特征和预后的关系及其与EZH2的相关性。结果：AML患者骨髓细胞中DNMT1mRNA的表达显著高于正常供者[（2.72±0.73）∶（0.89±0.27）,P〈0.01];EZH2mRNA的表达水平也显著高于正常供者[（4.39±1.06）∶（1.87±0.33）,P〈0.01];EZH2与DNMT1的表达水平呈显著正相关（r=0.51,P=0.002）;DNMT1mRNA表达水平与外周血幼稚细胞比例≥60%、WBC≥50×10^9/L显著相关（均P〈0.05）;DNMT1mRNA高表达组患者中位生存时间15个月（95%CI9～19个月）,显著低于DNMT1mRNA低表达组患者的32个月（95%CI27～40个月）（P=0.006）。结论：DNMT1和EZH2在AML患者中均高表达,二者表达水平呈正相关,DNMT1与AML患者预后不良相关。Objective：To investigate the relationship between DNMT1 and EZH2gene expression and clinical significant in patients with acute myeloid leukemia（AML）.Method：The expressions of DNMT1 and EZH2mRNA were quantified by RT-PCR in 50 cases of AML and 30 healthy controls.The relationship between DNMT1 and EZH2and clinicopathological factors and the prognosis were analyzed.Result：The expression of DNMT1 mRNA in AML was significantly increased as compared with that in healthy controls[（2.72±0.73）vs.（0.89±0.27）,P〈0.01].The expression of EZH2 mRNA in AML was also significantly increased compared to that in healthy controls[（4.39±1.06）vs.（1.87±0.33）,P〈0.01].The expression of DNMT1 was positively associated with EZH2（r=0.51,P=0.002）.Moreover,the expression of DNMT1 mRNA was associated with rate of blast cells in peripheral≥60% and WBC≥50×10^9/L（P〈0.05）.The overall survival of DNMT1 mRNA high expression group was 15 months（95%CI 9to 19 months）,which significant shorter than DNMT1 mRNA low expression group（32months,95%CI27to 40months;P=0.006）.Conclusion：The expressions of DNMT1 and EZH2are upregulated and associated with poor prognosis in AML.The expression of DNMT1 is positively associated with EZH2.
