摘  要:目的:通过模拟不同剂量分割方式照射食管癌细胞Eca109观察比较各组别细胞死亡方式及细胞毒效应的差异,尝试为临床食管癌非常规分割放疗提供潜在的理论依据。方法:设计具有相同BED的4种剂量分割模式:超分割组(1)、常规分割组(2)、大分割组(3)、大分割组(7),照射处于指数生长期的Eca109细胞后,进行Hoechst 33258免疫荧光染色观察细胞形态,通过CCK-8实验绘制细胞存活曲线。结果:Hoechst 33258染色观察细胞形态学变化:超分割组(1)受照射细胞中54.9%呈"凋亡样坏死"状态;常规分割组(2)50.5%的细胞呈"胀亡样坏死"状态,而30.3%的细胞呈凋亡样坏死状态;大分割组(3)受照射细胞中72.1%呈胀亡样坏死状态,19.2%呈凋亡样坏死状态;大分割组(7)受照射细胞中53.6%呈胀亡样坏死状态,7.1%呈凋亡样坏死状态。CCK-8检测各组细胞吸光度值(OD值),绘制存活曲线,常规分割组(2)、大分割组(3)与大分割组(7)在模拟照射结束后第1、4、7、10天进行比较,吸光度值均无显著性差异(P>0.05)。超分割组(1)在第1、4、10天吸光度值均显著低于其他模拟照射组(P<0.05)。在第7天时,各模拟照射组吸光度值均无显著性差异(P>0.05)。结论:超分割照射Eca109细胞大部表现为凋亡样坏死,大分割照射大部分表现为胀亡样坏死。在食管癌的治疗中,超分割放疗较其他剂量分割模式有更好的肿瘤控制率可能与这种死亡方式的差异有关。Objective:In hope to provide some theoretical basis for unconventional radiotherapy,we observe and compare cells' toxicity and death way between groups by simulating different dose fractionation on esophageal cancer cells Ecal09. Methods: Design the dose fraetionation mode with the same BED : the hyperfractionated group ( 1 ) , the conventional fractionation (2) , the hypofraetionated group (3) ,the hypofractionated group (7). After irradiation in Ecal09 cells in the exponential growth phase, the cell morphology was observed by Hoechst 33258 immunofluorescence staining,draw the cell survival curve by CCK-8 experiment. Results:Morphological changes of each group' s cell were observed in Hoechst 33258 staining. The hyperfractionated group ( 1 ) 54.9% of the irradiated cells were in the state of "apoptoticnecrosis". The conventional fractionation group (2) 50.5 % of the irradiated cells were in the state of "oncosticnecrosis" ;30.3% of the irradiated cells were in the state of "apoptoticnccrosis". The hypofractionated group ( 3 ) 72.1% of the irradiated cells were in the state of "oncosisnecrosis", 19.2% were in the state of "apoptoticnecrosis". The hypofractionated group (7) 53.6% of the irradiated ceils were in the state of "oncosisrtecrosis" ,7.1% were in the state of "apoptoticnecrosis". Absorbance values (OD) of cells in each group were detected with CCK-8, and the survival curve was drawn. By comparing between conventional fractionation group ( 2 ) , hypofraetionated group ( 3 ) and hypofraetionated group ( 7 ) on the lth,4th,Tth, 10th days after the simulation irradiation, the absorbance values of cells were not significantly different (P 〉 0.05 ). On the lth ,4th, lOtb day, the hyperfractionated Group ( 1 ) ' s absorbancc values were significantly lower than the other groups of simulation irradiation ( P 〈 0.05 ).
