摘  要:目的观察微小RNA-424-3p和5p在LNCaP中的表达,并比较其对LNCaP生物学行为的影响。方法应用实时荧光定量聚合酶链式反应(FQ-PCR)检测miR-424-3p和5p的表达量。利用细胞计数盒,平板克隆试验,Transwell小室,流式细胞仪检测LNCaP的增殖、迁移及细胞周期变化。结果 miR-424-3p和5p在LNCaP中的表达量高于RWPE-1;miR.-424-3p和5p下调后,LNCaP的增殖、迁移受到抑制,早期凋亡细胞比例增加,S期细胞比例增加。结论miR-424-3p和5p在LNCaP中高表达且5p升高更为显著,但比较两者对LNCaP生物学行为影响的大小,差异无统计学意义(P>0.05),提示miR-3p和5p对肿瘤细胞的生物学行为影响大小与其丰度并不一定成正比例关系。Objective To comparatively analyze the expressions of microRNA-424-3p and 5p in LNCaP cells, and detect their effects on biological behaviors of LNCaP cells. Methods Real-time fluorescence quantitative polymerase chain reaction (FQ-PCR) was used to detect relative expression of miR-424-3p and 5p in the RWPE-1 and LNCaP cells. CCK-8, flat plate cloning test and Transwell chamber assay were used to detect cell proliferation and invasion ability. Flow cytometry was used to analyzet cell apoptosis and cell cycle. Results The expressions of miR-424-3p and 5p in LNCaP cells were all higher than that in RWPE-1 cells. After MiR-424-3p and 5p inhibitors were transfected respectively, cell proliferation and migration were inhibited significantly, whereas the percentage of early apoptotic cells and S phase cell were all increased. Conclusion miR-424-3p and 5p highly expressed in LNCaP cells and miR-424 3P increased more significantly, but there were no differences in biological behavior changes of LNCaP cells between miR-424-3p and 5p (P〉0.05), suggesting their biological behavior changes are independent of expression level of miR-3p and 5p.
