去氨加压素和垂体后叶素对肝星状细胞增殖、收缩及分泌的影响

摘　　要：目的探讨醋酸去氨加压素和垂体后叶素对肝星状细胞（HSC）LX-2增殖、收缩及分泌的影响。方法以人HSC为研究对象，设置空白对照组、醋酸去氨加压素组和垂体后叶素组；两药物又分别设低、中、高浓度组。用WST-1检测HSC增殖情况；用倒置相差显微镜观察细胞形态和拍照后方格纸计算胶原凝胶板面积比法检测HSC收缩情况；用酶联免疫吸附法检测HSC分泌情况。对数据进行单因素方差分析。结果（1）醋酸去氨加压素低、中、高浓度组及垂体后叶素低、中、高浓度组的wsT～1A450值分别为0．459±0．017、0．467±0．024、0．436土0．015及0．495±0．011、0．507±0．015、0．501±0．009，与对照组相比（0．458±0．015），高浓度去氨加压素能抑制HSC增殖（P〈0．05）；各浓度垂体后叶素均明显促进HSC增殖沪值均〈0．05）。（2）各组胶原凝胶板面积比值分别为77．07±4．42、75．85±3．70、72．74±3．92及57．83±3．96、50．28±6．69、43．56±7．68，与对照组相比（77．40±4．56），垂体后叶素各浓度均能明显减小其面积（尸值均〈0．01）。（3）各组基质金属蛋白酶（MMP心浓度（ng／ml）分别为13．321±0．098、12．230±0．153、12．061±0．126及12．899±0．150、13．662±0．152、13．698±0．119，与对照组（12．281±0．169）相比，低浓度去氨加压素能增加MMP-2浓度（P〈0．01），而高浓度则降低其浓度妒〈0．05）；垂体后叶素各浓度均能显著增加MMP-2浓度（P值均〈0．01）。各组转化生长因子（TGF）p1浓度（pg／ml）分别为5．233±0．102、17．749±0．188、36.060±0．227及15．615±0．099、38．460±0．209、49．053±0．115，与对照组相比（0．751±0．083），去氨加压素和垂体后叶素均可促进HSC分泌TGFp1（P值均〈0．01），且呈浓度依赖关系。两种药物对应浓度组相比较，垂体后叶素的作用更强。与对照组�Objective To investigate the effects of desmopressin acetate and pituitrin on the proliferation, contraction, and secretion of hepatic stellate cells （HSCs）. Methods The human HSC cell line LX-2 was selected as the research model. And three groups were designed： blank control group, desmopressin acetate group （three subgroups： 1 ×10-10 tool/L, 1 × 10^-9 mol/L, and 1 × 10-8 mol/L dcsmopressin acetate）, and pituitrin group （three subgroups： 0.1 U/L, 1.0 U/L, and 10.0 U/L pituitrin）. Water-soluble tetrazolium salt （WST）- 1 assay was used to evaluate cell proliferation; collagen gel contraction assay was used to assess cell contraction; enzyme-linked imrnunosorbent assay （ELISA） was used to identify cell secretion. The data was subjected to one-way analysis of variance. Results （1） The results of WST-1 assay showed that the values of A450 in three desmopressin acetate subgroups （1 ~ 10-1~ mol/L, 1 ~ 10-9 mol/L, and 1× 10-8 tool/L） were 0.459±0.017, 0.467±0.024, and 0.436±0.015, respectively. And the values of A450 in three pituitrin subgroups （0.1 U/L, 1.0 U/L, and 10.0 U/L） were 0.495±0.011, 0.507±0.015, and 0.501±0.009, respectively. Compared with the control group, the desmopressin acetate at high concentration significantly inhibited the cell proliferation （P 〈 0.05）, but the pituitrin at three different concentrations significantly promoted the cell proliferation （P 〈 0.05）. （2） The collagen gel area ratios in three desmopressin acetate subgroups （1 × 10-10 mol/L, 1 ×10-9 mol/L, and 1 × 10-8 mol/L） were 77.07±4.42, 75.85±3.70, and 72.74±3.92, respectively. And the collagen gel area ratios in three pituitrin subgroups （0.1 U/L, 1.0 U/L, and 10.0 U/L） were 57.83±3.96, 50.28±6.69, and 43.56±7.68, respectively. Compared with the control group, the pituitrin at three different concentrations significantly reduced the collagen gel area （P 〈 0.01）. （3） The matrix metalloproteinase （MMP）-2 conceninations in r-tree desmopressin
