摘  要:目的以大鼠心肌缺血模型为对象,研究中药川芎对大鼠心肌缺血保护作用谱效关系及药效物质基础。方法建立中药川芎液相指纹图谱分析方法 ,以结扎大鼠冠状动脉致急性心肌缺血模型,运用多元回归分析、双变量相关分析,将川芎提取物药效数据与指纹图谱的共有峰的相对峰面积相关联,分析药物治疗心肌缺血作用的物质基础和谱效关系。结果藁苯内酯、川芎内酯、阿魏酸、川芎嗪均与药效呈显著正相关,其中阿魏酸、川芎嗪可显著降低血清中丙二醛(MDA)等的升高,可显著升高血清超氧化物歧化酶(SOD)活力,均为中药川芎治疗心肌缺血的主要有效成分。结论通过谱效关系研究获得了药效活性物质,建立了川芎药材谱效关系评价的方法 ,客观反映了药物内在质量,为该类中药的创新研究提供了借鉴。Objective To investigate the protective efficacy of Ligusticum Chuanxiong Hort. on myocardial ischemia injury in rats and to study the spectrum activity relationship of Ligusticum Chuanxiong Hort. Methods The Ligusticum Chuanxiong Hort. HPLC fingerprint analytical method was established, and the rat model of acute myocardial ischemia was established by applying left anterior descending coronary ligation. Bivariate correlation analysis and multivariate regression analysis were used to correlate the spectrum-activity relation- ship between fingerprints and anti- myocardial ischemia activity, and determine the spectrum activity relationship and efficacy material foundation of Ligusticum Chuanxiong Hort. Results Ligustilide, Chuanxiong lactone, ferulic acid, both TMP was a significant positive cor- relation with efficacy, ferulic acid, TMP can significantly reduce elevated serum MDA, etc.,ferulic acid, and ligustrazine showed a significant positive correlation with efficacy, in which the ferulie acid, and ligustrazine could significantly reduce elevated serum MDA, and sig- nificantly increase the activity of SOD, and were the main active ingredients in Ligusticum Chuanxiong Hort. on myocardial ischemia in- jury. Conclusion By Activity Relationship spectrum, the pharmacodynamie active substance is obtained, and the method of Chuanxiang spectrum activity relationship evaluation is established to objectively reflect the intrinsic quality of the drug, and provide references for the innovation study of such Chinese medicine.
