摘 要:目的:观察康艾注射液在体外对结肠癌 Lovo 细胞凋亡和增殖的影响。方法采用 Sub-G1法、Annexin-V /PI 法检测康艾注射液体外对结肠癌 Lovo 细胞凋亡的影响;Real-Time PCR(荧光定量 PCR)法检测康艾注射液体外作用结肠癌 Lovo 细胞后 Caspase-3、Bcl-2在 mRNA 表达水平的变化;流式细胞仪法检测康艾注射液体外对结肠癌 Lovo 细胞作用后增殖蛋白 Ki-67的表达。结果康艾注射液180μL/mL 处理 Lovo 细胞48 h 后,凋亡细胞比例 Sub-G1法对照组为(4.75±1.04)%,试验组为(45.61±5.22)%(t =7.68,P <0.001),Annexin-V /PI 法对照组凋亡细胞比例为(7.28±1.99)%,试验组为57.02±3.88)%,两组差异有统计学意义(t =11.42,P <0.01);Caspase-3表达增加(t =4.06,P <0.05),Bcl-2表达降低(t =5.69,P <0.01),两组差异有统计学意义;增殖蛋白 Ki-67对照组阳性表达率为(95.79±1.66)%,试验组为(65.84±4.80)%,两组差异有统计学意义(t =5.90,P <0.01)。结论康艾注射液在体外能够有效促进结肠癌 Lovo 细胞的凋亡,抑制其增殖。Objective To investigate the effects of KangAi injection on apoptosis and proliferation of rectal cancer Lovo cells in vitro.Methods SubG1 and Annexin -V /PI were used to examine the apoptosis of Lovo cells in vitro.Real -time PCR was used to examine the expression of Caspase -3 and Bcl -2 at mRNA level.Proliferation protein ki -67 was detected by flow cytometry.Results Lovo cells treated with KangAi injection 180μL/mL for 48h increased apoptosis rate from (4.75 ±1.04)% to (45.61 ±5.22)% with SubG1 (t =7.68,P 〈0.01 ),and (7.28 ±1.99)% to (57.02 ±3.88)% with Annexin -V /PI(t =11.42,P 〈0.01).Lovo cells treated with KangAi injection 180μL/mL for 48h increased the expression of Caspase -3 at mRNA level(t =4.06,P 〈0.05),while Bcl-2 decreased(t =5.69,P 〈0.01 ).Lovo cells treated with KangAi injection 180μL/mL for 48h decreased the expression of ki -67 from (95.79 ±1.66)% to (65.84 ±4.80)%(t =5.90,P 〈0.01 ).Conclusion KangAi injection can promote apoptosis and inhibit proliferation of rectal cancer Lovo cells in vitro.
