摘 要:目的:探讨海马区Fas蛋白表达与糖尿病大鼠局灶性脑缺血再灌注损伤的相关性。方法:选取健康雄性大鼠30只,以随机数字表法分为正常对照组、脑缺血再灌注组、糖尿病脑缺血再灌注组各10只。采用线栓法构造大鼠脑中动脉闭塞模型,并采用苏木精-伊红染色法观察海马CA1神经元缺失情况,采用免疫组化法检测Fas蛋白表达量。结果:苏木精-伊红染色结果中,正常对照组未观察到细胞凋亡和神经元缺失,而脑缺血再灌注组和糖尿病脑缺血再灌注组大鼠均观察到细胞凋亡和神经元缺失,其中糖尿病脑缺血再灌注组大鼠神经元缺失较为严重;正常对照组仅观察到极少量的Fas染色阳性细胞,而糖尿病脑缺血再灌注组和脑缺血再灌注组大鼠可以较为明显地观察到Fas染色阳性细胞,且糖尿病脑缺血再灌注组大鼠Fas蛋白表达显著高于脑缺血再灌注组。结论:糖尿病局灶性脑缺血再灌注损伤大鼠海马区神经元Fas蛋白的表达较为明显,Fas蛋白介导的细胞凋亡是海马神经元损伤的重要机制之一。OBJECTIVE: To probe into the correlation of hippocampus Fas protein expression and diabetes focal cerebral ischemia reperfusion injury in rats. METHODS: 30 healthy male rats were randomly selected and divided into normal control group( n = 10),cerebral ischemia reperfusion group( n = 10),diabetes cerebral ischemia reperfusion group( n = 10). MCAO was established by thread approach,CA1 neuronal loss was observed by HE staining,and Fas protein expression was detected by immunohistochemistry. RESULTS: HE staining results in the normal control group showed no apoptosis and neuronal loss,and cerebral ischemia reperfusion group and diabetes cerebral ischemia reperfusion group were observed apoptosis and neuronal loss,and the neuronal loss in diabetic cerebral ischemia reperfusion group were relatively severe. A small amount of Fas staining positive cells were observed in normal control group,yet Fas staining positive cells cerebral were clearly observed in ischemia reperfusion group and cerebral ischemia reperfusion group; and the Fas protein expression in diabetic cerebral ischemia reperfusion group was higher than that in cerebral ischemia reperfusion group. CONCLUSIONS: The diabetic rat focal cerebral ischemia reperfusion injury in the hippocampus neuron Fas protein expression is relatively apparent,Fas protein mediated apoptosis is one of the important mechanisms of hippocampal neuron damage.
