摘 要:目的:探讨微小RNA(mi R)-625-3p在结肠癌组织和细胞中的表达,了解其作用机制。方法:利用q RT-PCR分析结肠癌细胞株、癌组织和癌旁组织中mi R-625-3p的表达水平,探讨mi R-625-3p表达水平与结肠癌临床病理参数之间的关系;利用碘化丙啶染色和流式细胞术检测细胞凋亡和细胞周期变化;Western blot法检测mi R-625-3p对结肠癌细胞株凋亡相关蛋白的影响,初步了解其发挥影响的可能机制。结果:结肠癌组织mi R-625-3p的表达量比癌旁组织高(P<0.05),mi R-625-3p的表达量与结肠癌浸润深度、TNM分期及有无远处转移关系密切(P<0.05)。mi R-625-3p在结肠癌SW620细胞中的表达水平高于在SW480细胞中的表达。抑制mi R-625-3p的表达能显著抑制结肠癌细胞的周期(P<0.05)和促进凋亡;转染mi R-625-3p后,Bcl-2的表达量增加(P<0.05),Bax表达量没有显著变化。结论:mi R-625-3p能促进结肠癌细胞增殖,抑制结肠癌细胞凋亡,可能是结肠癌新的抗癌靶点。AIM: To investigate the expression of micro RNA-625-3p( mi R-625-3p) in colorectal carcinoma(CRC) and its underlying mechanism. METHODS: Quantitative real-time PCR was employed to detect the levels of mi R-625-3p expression in different CRC cell lines,CRC tissues and pair-matched adjacent normal tissues. The relationships between the expression levels of mi R-625-3p and the patients' clinicopathological parameters were estimated. The effects of mi R-625-3p on the apoptosis and the cell mitotic cycle of CRC cells were analyzed with propidium iodide staining and flow cytometry. The effect of mi R-625-3p on the apoptosis-related proteins was analyzed by Western blot. RESULTS: The expression level of mi R-625-3p in the CRC tissues was higher than that in the pair-matched adjacent normal tissues( P 0. 05). The expression of mi R-625-3p in the CRC tumor tissues was significantly correlated with the tumor infiltrative depth,TNM stage and distant metastasis( P 0. 05). The expression levels of mi R-625-3p in CRC SW620 cells were higher than that in SW480 cells. The CRC cell mitotic cycle was significantly inhibited and cell apoptosis was significantly promoted when the expression of mi R-625-3p was inhibited( P 0. 05). The expression of Bax protein didn't change and the expression of Bcl-2 protein increased after mi R-625-3p mimics were transfected into CRC SW620 cells( P 0. 05). CONCLUSION: mi R-625-3p may be a promising approach for the treatment of CRC by promoting cell proliferation and inhibiting apoptosis.
