摘 要:目的:探讨miR-145高甲基化在非小细胞肺癌(non-small cell lungcancer,NSCLC)组织中检测的意义。方法:收集2012年5月至2015年1月在我院住院治疗的75例NSCLC组织及癌旁组织,亚硫酸氢盐测序法PCR检测miR-145启动子甲基化状态,原位杂交法检测miR-145的表达,统计分析miR-145甲基化与患者临床参数的的关系及miR-145启动子甲基化与表达的关系。结果:miR-145在NSCLC组织中高甲基化的比例高于癌旁组织;miR-145高甲基化与NSCLC患者性别、年龄无关,与组织分化程度密切相关;miR-145甲基化状态与NSCLC组织中miR-145表达呈负相关。结论:miR-145高甲基化与NSCLC分化程度密切相关,可能miR-145甲基化检测可作为NSCLC分化程度判定的潜在分子生物学标记物。Objective To explore the significance of detecting miR-145 hypermethylation in non-small cell lung cancer(NSCLC). Methods Seventy-five NSCLC tissues and adjacent tissues was collected from May 2012 to January 2015. The methylation status of miR-145 promoter was tested by bisulfite sequencing PCR(BSP) and the expression of miR-145 evaluated by ISH. The correlation between miR-145 methylation and clinical parameters, and the relationship between miR-145 methylation and expression were statistically analyzed. Results The hypermethylation ratio of miR-145 in NSCLC tissues was higher than that in the adjacent tissues. Hypermethylation of miR-145 was correlated with NSCLC differentiation stage, but not with gender and age. In NSCLC, miR-145 hypermethylation was negatively related with its expression. Conclusion Mi R-145 hypermethylation is closely related with differentiation stage in NSCLC tissues. Mi R-145 hypermethylation may be a potential biomarker for estimating NSCLC differentiation stage.
