摘 要:目的:鉴定华佗再造丸中具有细胞保护作用的化合物。方法:采用PC12细胞和小鼠脑微血管内皮细胞(b End.3)氧化损伤和缺糖缺氧再灌注(OGR)损伤模型测定,从中得到的72种单体成分中具有细胞保护活性和促神经细胞增殖活性的化合物。结果:可提高氧化损伤的PC12细胞存活率的化合物有槲皮素和异鼠李素;可提高OGR损伤的PC12细胞存活率的单体有异甘草素、3-咖啡酰奎尼酸和人参皂苷Rb1。可促进正常PC12细胞增殖的单体化合物有人参皂苷Rg2、胆酸钠等8种化合物。可提高氧化损伤的b End.3细胞存活率的单体有槲皮素、芍药苷等6种化合物。可提高OGR损伤的b End.3细胞存活率的单体有槲皮素、人参皂苷Rg1等6种化合物。结论:除槲皮素对不同细胞不同损伤模型细胞均有保护作用,为广谱细胞保护剂外,其余化合物均有一定的细胞选择性或模型选择性保护作用,对神经细胞增殖有促进作用的化合物未见有细胞保护作用。这些结果为华佗再造丸治疗脑血管疾病提供了进一步的药理学依据。AIM: To indentify the compounds with cytoprotective activity that were isolated from Huatuozaizao pill. METHODS: PC12 models injured by H202 or Oxygen-Glucose Deprivation (OGD) and mouse brain microvascular endothelial cell line (bEnd. 3 ) model injured by H202 or OGD were used to detect the cytoprotective activity of 72 compounds isolate from Huatuozaizao pill and their proliferation promoting activity. RESULTS : The vi- ability of PC12 injured by oxidation was increased by quercetin and isorhamnetin; the viability of PC12 in- jured by OGD was increased by isoliquiritigenin, 3- caffeoylquinic acid and ginsenoside Rbl; PC12 pro- liferation was promoted by Ginsenoside Rg2, sodium cholate, wild astragalus glycoside, strychnine, as- tragalin , hyodesoxycholic acid, ginsenoside Rg3 and albiflorinstd; the viability of bEnd. 3 injured by H202 was increased by quercetin, peoniflorin, gin- senoside Rgl, R-ginsenoside Rhl-isomer, ginsen-oside Rd, ginsenoside R4; the viability of bEnd. 3 injured by OGD was increased by qercetin, ginsen- oside Rgl, R-ginsenoside Rhl-isomer, ginsenoside Rd, 4-caffeoylquinic acid and 5-caffeoylquinic acid. CONCLUSION: Besides the broad-spectrum cyto- protective acivity of qercetin, other compounds also show cell selective or injury model selective cytopro- tective activity ; compounds promoting PC12 prolifer- ation show no cytoprotective activity. Our research exhibits the evidence of the compounds with cytopro- tective activity and the compounds with promoting PC12 proliferation in Huatuozaizao pill, which could be referential for studying the mechanism of Huat- uozaizao pill in treating cerebrovascular disease.
