摘  要:目的比较去甲斑蝥素(noreantharidin,NCTD)单独用药和联合ABT-737 2种用药方式对宫颈癌细胞增殖及凋亡的影响。方法应用四甲基偶氮唑盐(MTT)法,检测NCTD单药或联合ABT-737 2种用药方式对宫颈癌细胞株He La和Si Ha增殖的影响;应用流式细胞术分析NCTD联合ABT-737对宫颈癌细胞凋亡的影响。结果 NCTD单独用药能够显著抑制宫颈癌细胞株He La和Si Ha的增殖(P<0.05),且抑制作用呈剂量依赖性,而且NCTD与ABT-737联用后对宫颈癌细胞增殖的抑制作用明显增强。NCTD和ABT-737联合用药的促凋亡作用比单独用药效果显著增强(P<0.05)。结论体外单独使用NCTD对宫颈癌细胞He La和Si Ha的增殖具有明显抑制作用,联合使用ABT-737后对其增殖抑制以及促凋亡作用更明显。Objective To study the influence of NCTD (noreantharidin) and ABT-737 on the proliferation and apoptosis of cervical cancer cells. Methods MTT method was used to detect the influence of NCTD single-agent or joint ABT-737 on the proliferation of HeLa and SiHa cervical cancer cell lines;flow cytometry was used to analyze the influence of NCTD joint ABT-737 on the apoptosis of cervical cancer cells. Results NCTD single-agent can significantly inhibit the proliferation of HeLa and SiHa cervical cancer cell line( P 〈 0.05 ) , and the inhibitory effect was dose dependent. NCTD inhibition was enhanced after combina- tion with ABT-737. Combination of NCTD and ABT-737 had more significant effect on promoting apoptosis than any single agent ( P 〈 0.05 ). Conclusion The proliferation of SiHa and HeLa in cervical cancer cells is significantly inhibited by NCTD in vitro, the inhibition effect of ABT-737 and NCTD on proliferation and apoptosls is more obvious.
