摘  要:目的:探讨血清维生素D浓度与子宫内膜癌的相关性。方法:收集子宫内膜相关病变患者118例为试验组,其中子宫内膜增生症22例,子宫内膜癌96例,并以30例体检健康妇女作为对照组。收集所有研究对象的清晨空腹肘静脉血标本。采用放射免疫法检测标本血清中维生素D的浓度。结果:子宫内膜癌患者血清维生素D水平较正常对照组减低,随着子宫内膜癌病理分级及FIGO分期的升高,血清维生素D浓度逐渐减低,有淋巴转移患者较无淋巴转移者血清维生素D浓度亦减低,差异均有统计学意义。结论:血清维生素D浓度与子宫内膜癌恶性程度呈负相关。Objective To investigate the biological effects of MeCP2 on cervical cancer Hela cells and molecular mechanism.Methods siRNA interference were used to knockdown MeCP2.MTT assay were used to analyze the effects of MeCP2 on proliferation of cervical cancer Hela cells.To analyze the effects of MeCP2 on the cell cycle and apoptosis of cervical cancer Hela cells with flow cytometer.To examine the effects of MeCP2 siRNA on the expression of Cyclin D1,P-ERK1/2and P-p38 with Western Blot.Results MeCP2 mRNA and protein significantly decreased in MeCP2 siRNA group compared to negative control group(P〈0.01).MeCP2 siRNA inhibits proliferation of cervical cancer Hela cells.The number of S and G2/M phase cells significantly decreased in MeCP2 siRNA group compared to negative control group(P〈0.01),meanwhile the number of G1/G0 phase cells remarkably increased(P〈0.01).The number of early apoptotic cells and late apoptotic cells remarkably increased in MeCP2 siRNA group compared to negative control group(P〈0.01).It was found that Cyclin D1 and P-ERK1/2proteins significantly decreased in MeCP2 siRNA group compared to negative control group(P〈0.01),P-p38 proteins remarkably increased(P〈0.01).Conclusion MeCP2 promotes proliferation of cervical cancer Hela cells through regulating P-ERK1/2and Cyclin D1 expression,and inhibits apoptosis by suppressing P-p38 MAPK signal pathway.
