KLF4在宫颈癌中的表达及临床意义

摘　　要：目的:探讨转录因子KLF4在宫颈癌中的表达及临床意义。方法:采用免疫组化SP法检测80例宫颈癌、30例宫颈原位癌、30例正常宫颈组织中KLF4蛋白的表达情况并分析其与宫颈癌临床病理因素之间的关系;蛋白质免疫印迹法检测30例新鲜宫颈癌组织及配对宫颈癌旁组织的蛋白表达水平。结果:免疫组化结果显示KLF4在宫颈癌、宫颈原位癌及正常宫颈组织中的阳性表达率分别为11.25%、56.67%和86.67%,在宫颈癌中的阳性表达较其它各组明显下降(P<0.05)。KLF4在宫颈癌组织中的表达与患者年龄、肿瘤分化程度、组织学分型无关(P>0.05),随着临床分期的进展,KLF4阳性表达率呈下降趋势(P<0.05);蛋白质免疫印迹检测结果提示KLF4在宫颈癌中的表达水平低于癌旁组织,差异具有统计学意义(P<0.05)。结论:KLF4在宫颈癌中的表达水平与其进展呈负相关,在宫颈癌发生发展的过程中发挥抑癌基因作用,有望成为预测宫颈癌发生发展、靶向治疗及判断患者预后的新的参考指标。Objective： To investigate the expression and clinical significance of Krüppel- like factor 4 in cervical cancer. Methods： The expression of KLF4 protein in 80 cases of cervical cancer,30 cases of cervical cancer insitu and30 cases of normal cervical tissues was examined by SP immunohistochemical staining and the significance of the expression with clinicopathologic parameters was analyzed. Western Blot analyzed the expression level of 30 pairs tumor and tumor adjacent tissues. Results： The positive expression of KLF4 in cervical cancer,cervical cancer in situ and normal cervical tissues was 11. 25%,56. 67% and 86. 67% respectively. KLF4 expression in cervical cancer significantly decreased compared with other groups（ P〈0. 05）. There was no difference between the patient＇s age,histological type and differentiation（ P〉0. 05）,while obvious difference was related to FIGO stage in cervical cancer（ P〈0. 05）. Western Blot analysis showed that KLF4 expression was markly lower in the cervical cancer tissues compared to their adjacent normal cervical tissues. Conclusion： The expression of KLF4 in cervical cancer was negatively correlated with tumor progress and it plays a role of tumor suppressor genes in the development of cervical cancer,which was expected to be a new marker to predict the occurrence and the prognosis of patients with cervical cancer.
