摘 要:目的:观察脾虚小鼠感染白色念珠菌后,肠道病理学和超微结构的改变,为临床脾虚证的防治提供病理学依据。方法:健康SPF级昆明种小白鼠40只,随机分为两组:空白组(N组,20只)及脾虚模型组(M组,20只)。对脾虚模型组小鼠采用饮食不节加劳倦过度的方法复制脾虚小鼠模型。模型复制成功后,再次分别将N组及M组小鼠各随机分为两组:空白对照组(N1组,10只)、空白感染白色念珠菌组(N2组,10只)、脾虚模型对照组(M1,10只)、脾虚感染白色念珠菌组(M2,10只)。脾虚模型复制成功的次日对N2组和M2组小鼠经口感染白色念珠菌,白色念珠菌浓度为2×108 CFU/m L,剂量为0.2 m L/10 g体重,N1、M1组小鼠给予等量生理盐水经口灌胃。染菌第14天,处死各组小鼠,采用HE染色的方法观察各组小鼠小肠组织病理学改变,采用透射电镜的方法,观察各组小鼠小肠超微结构改变。结果:空白对照组小鼠小肠组织黏膜完整,绒毛排列整齐,肌层薄厚均匀适中。脾虚对照组小鼠与空白对照组比较,差别不明显,仅偶见小肠绒毛排列不均匀。空白感染白色念珠菌组和脾虚感染白色念珠菌组小鼠均显示出不同程度的病变,尤以脾虚感染白色念珠菌组小鼠病理改变尤为明显,主要表现为绒毛缺失,排列不整齐,黏膜下层有炎细胞浸润。空白对照组小鼠小肠微绒毛排列整齐,细胞内细胞器丰富,线粒体、内质网、核糖体清晰可见。脾虚对照组小鼠小肠微绒毛略短,排列比较整齐,与空白对照组比较,差异不显著。空白感染白色念珠菌组小鼠小肠微绒毛稀疏,长短不一,细胞质内线粒体肿胀、偶见空泡样改变。脾虚感染白色念珠菌组小鼠小肠微绒毛明显减少,排列紊乱,线粒体明显肿胀,嵴消失,呈空泡样改变,内质网扩张,表面核糖体脱落。结论:脾虚状态下的机体需时刻注意白色念珠菌感染的发生,预防白色念珠菌菌感染将是脾虚�Objective:To observe the spleen deficiency model mice infected by white candida albicans,the changes of intestine pathology and ultrastructure,for clinical prevention and cure of the spleen deficiency pathology basis. Methods:40 healthy SPF mice were randomly divided into two groups:control group(N group,20 mice);spleen deficiency model group(M group,20 mice). On the model of spleen deficiency mice model by replication of improper diet and overstrain method of replication of spleen deficiency mice. After successful model replication,M and N mice were randomly divided into two groups:control group(N1 group,10 rats);control infected with candida albicans group(N2,10);spleen deficiency model group(M1,10);spleen deficiency infected with candida albicans group(M2,10). N2 and M2 mice were infected with candida albicans,and the concentration of white candida albicans was 2 ×108 CFU/m L,and the dose was 0.1 m L/10 g. N1 and M1 mice were given equal normal saline to fill the stomach through the mouth. After 14 days,to observe each mouse small intestine tissue pathology change,and observation group mice intestine ultrastructural changes. Results:Control group mice intestinal mucous membrane is integrity,and the villus is neatly;spleen deficient model group mice compared with control group,the difference is not obvious,only occasional small intestinal villus arranged unevenly. The control infected with candida albicans group and spleen deficient infected with candida albicans group mice showed different degrees of pathological changes,especially in the spleen deficient infected with candida albicans group,main show that intestinal villus is loss,not neatly,with inflammatory cells infiltration under intestinal mucous. Control group mice intestine microvilli neatly,abundant organelles in the cell,mitochondria,endoplasmic reticulum,ribosomes are visible.
