摘  要:目的观察BMSC对经基因改造的具有不同转移潜能肝癌的影响。方法 63只裸鼠随机分成8组:MHCC97-L组(9只)、MHCC97-L NC组(8只)、MHCC97-L OPN沉默组(8只)、MHCC97-L TGFβ1沉默组(5只)、MHCC97-H组(8只)、MHCC97-H NC组(8只)、MHCC97-H OPN沉默组(9只)、MHCC97-H TGFβ1沉默组(8只)。肿瘤体积50 mm3左右开始瘤内注射人间充质干细胞BMSC,1×10^5/只,每周2次,连续4周。计算瘤体积抑制率(%)和瘤重抑制率(%),BMSC对经基因沉默的肝癌动物模型干预结果进行病理检测。结果与MHCC97-L组比,BMSC对MHCC97-L NC组瘤体积和瘤重均有促进作用,与MHCC97-L组比,BMSC对MHCC97-L OPN沉默组瘤体积和瘤重均有明显的抑制作用,而BMSC对MHCC97-L TGFβ1沉默组瘤体积和瘤重均有促进作用。与MHCC97-H组比,BMSC对MHCC97-H NC组瘤体积和瘤重均有轻度抑制作用,与MHCC97-H组比,BMSC对MHCC97-H OPN沉默组和MHCC97-H TGFβ1沉默组瘤体积和瘤重均有显著的抑制作用。与MHCC97-H组及MHCC97-H NC组比较,MHCC97-H OPN沉默组纤维增生及核分裂象较少,而MHCC97-H TGFβ1沉默组纤维增生明显,核分裂象较多。与MHCC97-L比较,MHCC97-L NC、MHCC97-L OPN沉默组、MHCC97-L TGFβ1沉默组见较多坏死区、较多肿瘤细胞凋亡、坏死,并可见数个核分裂象。结论抑制肿瘤OPN表达BMSC可更好地抑制肿瘤的增殖,而抑制肿瘤TGFβ1表达BMSC并不能抑制肿瘤的增殖。相应的抗体沉默OPN可能是治疗肿瘤的一个比较好的靶点,配合BMSC生物治疗会提高治疗的生物效能。Objective To observe the effect of BMSC on hepatocellular carcinoma( HCC) with different metastatic potential by gene interference. Methods Sixty-three nude mice were randomly divided into eight groups,including MHCC97-L blank control group( 9 nude mice),MHCC97-L gene negative control group( 8 nude mice),MHCC97-L OPN gene interference group( 8 nude mice),MHCC97-L TGFβ1 gene interference group( 5 nude mice) and MHCC97-H blank control group( 8 nude mice),MHCC97-H gene negative control group( 8 nude mice),MHCC97-H OPN gene interference group( 9 nude mice),MHCC97-L TGFβ1 gene interference group( 8 nude mice). When tumor volume grown to 50 mm3,human mesenchymal stem cells( BMSC) was injected,and 1 × 10^5/ one,two times a week,for 4weeks. Tumor volume inhibition rate( %) and tumor weight inhibition rate( %) were calculated,and pathological test was used for HCC animal model with gene silencing. Results Compared with the MHCC97-L blank control group,BMSC could promote the tumor volume and tumor weight of MHCC97-L NC group,and could inhibit tumor volume and tumor weight of MHCC97-L OPN gene interference group. However,BMSC could promote the tumor volume and tumor weight of MHCC97-L TGFβ1 gene interference group. Compared with the MHCC97-H blank control group,BMSC could mildly inhibit the tumor and tumor weight of MHCC97-H gene negative control group,and could inhibit tumor volume and tumor weight of MHCC97-H OPN and TGFβ1 gene interference groups. Compared with MHCC97-H blank control group and MHCC97-H gene negative control group,the fiber proliferation and nuclear division of MHCC97-H OPN gene interference group were less,while the MHCC97-H TGFβ1 gene interference group had obvious fiber hyperplasia and more nuclear division. Compared with MHCC97-L blank control group,MHCC97-L gene negative control group,MHCC97-L OPN gene interference group and MHCC97-L TGFβ1 gene interference group could see more necrotic areas,more tumor cell apoptosis,necrosis,and can be seen a n
