摘 要:目的观察低氧预处理对长时间大鼠急性高眼压模型视网膜节细胞存活的作用。方法实验研究。对2011年2月至2012年12月在汕头大学·香港中文大学联合汕头国际眼科中心应用健康成年Fischer344雌性大鼠,8~12周龄,前房灌注生理盐水诱导急性高眼压模型(灌注高度1496mm,2h)作为急性高眼压组(n=5),低氧预处理(氧浓度11%,2h)联合急性高眼压为联合组(n=10),及正常对照组(n=10),前房灌注后7天及18天后进行视网膜节细胞计数;多组间比较采用单因素方差分析,两两比较采用Bonferroni检验。结果急性高眼压组及联合组前房灌注后7天的平均视网膜节细胞密度分别为(1815.0±84.9)mm^2,(1697.4±291.0)mm^2,18天分别为(1468.9±92.0)mm^2,(1373.4±87.0)mm^2。正常对照组平均视网膜节细胞密度为(2259.0±163.8)mm^2,急性高眼压组和联合组7天及18天组间视网膜节细胞皆差异无统计学意义(P=0.775,P=0.318),两组各时间点的平均视网膜节细胞密度与正常对照组间差异具有统计学意义(P〈0.05)。结论低氧预处理对延长缺血时间的大鼠急性高眼压的视网膜节细胞无显著保护作用。Objective To observe the effect of hypoxic preconditioning on the retinal ganglial cells (RGCs) survival in long time acute ocular hypertension rat model. Methods 25 healthy adult female Fischer344 rats were used for this study. The acute ocular hypertension group: right eye of rat was induced acute ocular hypertension (height of 1496 mm, 2 h, n=10); the combined group: hypoxic preconditioning (11% O2, 2 h) combined acute ocular hypertension (n=10); normal control group (n=5). RGCs of both groups were counted after 7 days and 18 days. Data were statistically analyzed by using one-way analysis of variance and Bonferroni test. Results The mean RGCs density of the acute ocular hypertension group and combined group after 7 days was (1815.0±84.9)mm^2, (1697.4±291.0) mm^2 respectively, and after 18 days was (1468.9±92.0) mm^2 and (1373.4±87.0) mm^2 respectively. There was no statistically significant difference between the two groups (P =0.775, P = 0.318). The mean RGCs density of normal control group was (2259.0±163.8) mm^2. Either experimental group had statistically significant difference with normal control group (P 〈0.05). Conclusion Hypoxic preconditioning has no significant effect on RGCs survival in acute ocular hypertension rat model with extend increased time of ischemia.
