摘 要:目的观察紫杉醇(PA)对人胃癌细胞株SGC7901放射敏感性的影响。方法采用MTT法获得PA对SGC7901细胞的半数抑制浓度(IC50),并以20%的IC50剂量为增敏浓度;克隆形成实验检测单纯照射组与PA(增敏浓度)+照射组在0、2、4、6、8 Gy X线照射后的细胞存活率,并观察细胞周期的改变及细胞形态的变化。结果 PA对SGC7901细胞的IC50为16.11μg/ml,随着照射剂量的逐渐增加,单纯照射组和PA+照射组细胞存活率呈下降趋势,从4 Gy开始,PA+照射组细胞的存活率低于单纯照射组(P<0.05);PA+照射组的G_2/M期细胞比例高于其他两组,S期细胞比例低于其他两组(P<0.05)。结论 PA对胃癌细胞株SGC7901具有放射增敏作用,其机制可能与诱导细胞凋亡及G_2/M期阻滞有关。Objective To observe the effect of paclitaxel (PA) on the radiosensitivity of human gastric cancer cell line SGC7901.Methods MTT assay was employed to obtain the 50% inhibition concentration (IC50) of PA on SGC7901 ceils. 20% of the IC50 dose was treated as sensitizing concentration.The clonogenic assay was used to analyze the cell viability of radiation group and PA (sensitizer concentration) plus radiation group with X-ray of 0,2,4,6 and 8 Gy.The changes of cell cycle and cell morphology changes were observed.Results The IC50 of PA on SGC7901 cells was 16.11 μg/ml. The cell viability got decreased with increasing radiation dose in radiation group and PA plus radiation group.The cell viabilities of PA plus radiation groupwere lower than those of radiation group at the dose of 4,6 and 8 Gy.There was higher percentage in the G2/M phase,and lower percentage in the S phase in PA plus radiation group versus the other two groups (P〈0.05).Conclusion PA can enhance the radiosensitivity of SGC7901 cells.The mechanism is connected with that it can induce cell apoptosis and G2/M phase retardation.
