摘 要:目的:探讨人参皂苷单体Rh2体外抑制鼻咽癌肿瘤干细胞增殖的作用以及对白细胞介素(IL)-6水平影响。方法:应用无血清悬浮培养法在鼻咽癌低分化细胞株CNE-2中筛选分离出鼻咽癌肿瘤干细胞样细胞亚群CNE-2S;按随机法将细胞分为生理盐水组、人参皂苷单体Rh2(30μmol/L)组和人参皂苷单体Rh2(60μmol/L)组;应用MTT法测各组CNE-2S细胞增殖;CNE-2S细胞凋亡应用DAPI染色法检测,分析细胞的光密度和面密度变化;酶联免疫吸附(Elisa)法测定各组CNE-2S细胞的IL-6水平。结果:30μmol/L人参皂苷单体Rh2对鼻咽癌肿瘤干细胞的抑制率为15.46%,明显低于生理盐水组(P<0.05);60μmol/L人参皂苷单体Rh2组的抑制率为44.33%,极显著低于生理盐水组(P<0.01)。人参皂苷单体Rh2干预后鼻咽癌肿瘤干细胞的DAPI染色光密度和面密度值均显著减小,比较差异有统计学意义(P<0.01);且60μmol/L人参皂苷单体Rh2组的光密度和面密度值均明显小于30μmol/L人参皂苷单体Rh2组,比较差异有统计学意义(P<0.01)。干预后3 d和6 d,人参皂苷单体Rh2组IL-6表达水平明显减少(P<0.01),且60μmol/L人参皂苷单体Rh2组的IL-6表达水平均分别明显低于30μmol/L人参皂苷单体Rh2组,比较差异有统计学意义(P<0.01)。结论:人参皂苷单体Rh2对体外培养鼻咽癌肿瘤干细胞的增殖具有明显抑制作用,且可促进其凋亡,下调IL-6表达可能在其中发挥重要作用。Objective: To discuss the inhibiting effect of ginsenoside Rh2 in vitro on nasopharynx cancer CSCs proliferation and on interleukin( IL)-6 levels. Methods: Nasopharynx cancer CSCs subgroup CNE-2S was screened and separated in poorly differentiated cell strain CNE-2 of nasopharynx cancer through serum-free suspension culture method; and then cells were randomly divided into normal saline( NS) group,ginsenoside Rh2( 30 μmol / L) group and ginsenoside Rh2( 60 μmol / L) group; MTT method was adopted for detecting the CNE-2S cell proliferation in each group; DAPI staining method was adopted for detecting the CNE-2S cell apoptosis,and cell optical density and surface density changes were analyzed; ELISA method was adopted for detecting the IL-6 levels of CNE-2S cells in each group. Results: Inhibition rate of ginsenoside Rh2( 30 μmol / L) to cancer stem cells of nasopharynx cancer was15. 46%,which was significantly lower than that in NS group( P〈0. 05); ginsenoside Rh2( 60 μmol / L) inhibition rate was 44.33%,which was extremely significantly lower than that in NS group( P〈0. 01). After ginsenoside Rh2 intervention,DAPI staining of cancer stem cell of nasopharynx cancer was significantly weakened,and optical density and surface density values were significantly decreased( P〈0. 01); optical density and surface density values in ginsenoside Rh2( 60 μmol / L) group were significantly lower than that in ginsenoside Rh2( 30 μmol / L) group( P〈0. 01). 3 d and 6 d after intervention,IL-6 expression levels were significantly reduced in ginsenoside Rh2 group( P〈0. 01),and IL-6 expression levels in ginsenoside Rh2( 60 μmol / L) group were significantly lower than that in ginsenoside Rh2( 30 μmol / L)( P〈0. 01). Conclusions: Ginsenoside Rh2 has significant inhibiting effects on nasopharynx cancer CSCs proliferation in vitro,in addition,it could promote the cell apoptosis and reduce the IL-6 expression.
