摘  要:以钠化蒙脱石、硝酸锌为原料,采用液相离子交换法制备载锌蒙脱石抗菌中间体;应用响应面法设计优化工艺、扫描电镜、XRD观察载锌蒙脱石形貌、分析组成;抑菌圈及最低抑菌浓度法(MIC)研究其抗菌性能;应用水热法,在镁合金AZ31表面制备载锌蒙脱石涂层,电镜观察其形貌。结果表明,采用Design-Expert软件中的Boxbehnken设计的二次多项式模型比较显著,得出最佳优化条件,硝酸锌为4mmol,温度为80℃,时间为4.5h,蒙脱石载锌量达到94.65mg/g;扫描电镜观察载锌蒙脱石颗粒细小,呈不规则形,很少聚集成团;XRD分析发现蒙脱石片层间距由原来的1.48nm增加到1.57nm;抗菌实验表明,载锌蒙脱石对大肠杆菌和金黄色葡萄球菌具有较好的抗菌活性,MIC为6 400mg/mL;电镜观察显示,载锌蒙脱石在AZ31镁合金表面形成比较致密的1层涂层,与基体结合紧密,厚度达42μm,说明载锌蒙脱石抗菌中间体可成功应用于镁合金表面。Zn-montmorillonites(Zn-MMT)as antibacterial compounds were prepared from natrium montmorillonite,zinc nitrate by an ion-exchange reaction.The effects of the amount of zinc nitrate,reaction temperature and reaction time on the zinc content of montmorillonite were optimized with response surface methodology.ZnMMT were characterized by means of XRD and SEM.The results of bacterial growth tests were confirmed by determination of zone of inhibition against E.coli,S.aureus and the minimum inhibition concentrations(MIC).The Zn-MMT coating on the magnesium alloy was prepared via hydrothermal method.The structure of ZnMMT coating was investigated by SEM.Design-expert 8.0was used to established the mathematical model.The results showed that quadratic model was the best model to describe the relationship between the zinc content of Zn-MMT and the three factors.With adding zinc nitrate 4mmol,reaction temperature of 80 ℃,reaction time of 4.5h,the zinc content of montmorillonite reached 94.65mg/g.SEM micrographs showed the powders are tiny particulate,irregular shape,seldom aggregation.The interlayer spacing of MMT expand from 1.48to1.57nm;antibacterial experiments showed that Zn-MMT provided powerful biocidal to S.aureus and E.coli,and the minimum inhibitory concentration was 6 400μg/mL.And then,using hydrothermal method,the surface of AZ31 alloy was successfully coated with Zn-MMT.
