摘 要:目的建立斑点免疫金渗滤技术检测抗心磷脂抗体(Ac Ab)的新方法。方法采用牛心磷脂抗原,以胶体金颗粒结合的羊抗人Ig G为标记抗体,根据斑点免疫金渗滤技术原理,建立斑点免疫金渗滤技术法检测人血清抗心磷脂抗体。分别检测78份抗心磷脂抗体阳性血清和80份正常体检女性血清,并与酶联免疫吸附试验(ELISA)的结果对比分析。结果与酶联免疫吸附试验的结果对比,其符合率为91.77%,特异性为95.00%,敏感性为88.46%,交叉试验与重复试验结果显示,斑点免疫金渗滤技术测定抗心磷脂抗体具有较好的稳定性和特异性。结论斑点免疫金渗滤技术可以快速准确地测定血清抗心磷脂抗体,便于推广普及。Objective To develop a new method for detection of anti-cardiolipin antibody(Ac Ab) with immune colloidal gold infiltration technology. Methods Using cattle cardiolipin antigen, colloidal gold particles conjugated goat antihuman Ig G antibody was labeled using the principles of immune colloidal gold infiltration technology, and then was used to build a new method for detecting anti-cardiolipin antibody in human serum. 78 positive serum samples of anticardiolipin antibody and 80 serum samples of normal physical examination female were detected, and the result was compared with enzyme-linked assay(ELISA) and immune colloidal gold infiltration technology. Results Compared with the result of ELISA, the consistency of immune colloidal gold infiltration technology was 91.77%. The specificity of the assay was 95.00% and sensitivity was 88.46%. The preparation was stable after 6 months refrigerator storage. Conclusion Immune colloidal gold infiltration technology can quickly and accurately measure serum anti-cardiolipin antibody, and should be promoted.
