摘  要:目的:研究前列腺癌组织及细胞株中丝氨酸-苏氨酸激酶11(liver kinase B1/serine-threonine kinase 11,LKB1)、转化生长因子-β1(transforming growth factor-β1,TGF-β1)的表达情况,探讨LKB1表达在前列腺癌发生演进中的作用。方法:用免疫组化SP法、Western blot和RT-PCR检测LKB1及TGF-β1在前列腺增生和前列腺癌组织,以及2种细胞株(正常前列腺上皮RWPE-1,前列腺癌细胞株PC-3)中的蛋白和m RNA表达情况。结果:前列腺癌组织中LKB1、TGF-β1的阳性表达率分别为55%、70%,前列腺增生组织中LKB1、TGF-β1的阳性表达率分别为83.3%、40%,前列腺癌组织中LKB1的表达显著低于前列腺增生组织(P<0.05),前列腺癌组织中TGF-β1的表达明显高于前列腺增生组织(P<0.05),LKB1和TGF-β1在前列腺癌中的表达呈负相关(r=-0.373,P=0.018);前列腺增生组织中LKB1的蛋白和m RNA表达水平明显高于前列腺癌组织(P<0.01),前列腺癌组织中TGF-β1的蛋白和m RNA表达水平显著高于前列腺增生组织。免疫细胞化学、Western blot、RT-PCR结果显示:LKB1在人前列腺上皮细胞株RWPE-1中的蛋白及m RNA表达水平明显高于前列腺癌细胞株PC-3(P<0.01);TGF-β1在前列腺癌细胞株PC-3中的蛋白及m RNA表达水平明显高于人前列腺上皮细胞株RWPE-1(P<0.01)。结论:LKB1在前列腺癌中表达下调,而TGF-β1在前列腺癌中过表达,LKB1在前列腺癌的发生过程中可能起着抑制TGF-β1的作用;对LKB1和TGF-β1相互作用机制的深入探讨有可能使其成为研究前列腺癌发生进展的分子机制新靶点。Objective:To observe the effect of liver kinase B1/serine-threonine kinase 11 (LKB1) on the progression of prostate cancer by studying the expression of LKB 1,transforming growth factor-β1 (TGF-β1) in prostate cancer tissues and cells. Methods:Two cell lines(normal prostatic epithelial cell line RWPE-1, prostatic carcinoma cell lines PC-3) were detected by SP method, RT-PCR and Western blot in order to detect the expression of LKB,TGF-β1 mRNA and protein on human prostatic hyperplasia and prostate can- cer tissues. Results:The positive rates of LKB1 and TGF-β1 were 55% and 70% in prostatic carcinoma,and 83.3% and 40% in prostatic hyperplasia. The positive rates of LKB1 were lower in prostatic carcinoma than in prostatic hyperplasia(P〈O.05), and the positive rates of TGF-β1were higher in prostatic carcinoma than in prostatic hyperplasia(P〈0.05). However,LKB1 expression had a negative correlation with TGF-β1 (r=-0.373 ,P=0.018). The relative levels of LKB1 protein and mRNA were higher in prostatic hyperplasia than in prostatic carcinoma (P〈0.01). However, the relative levels of TGF-β1 protein and mRNA were higher in prostatic carcinoma than in prostatic hyperplasia. The level of LKB1 protein and mRNA were higher in prostatic epithelial cell line RWPE-1 than in prostatie carcinoma cell line PC-3(P〈0.01 ) by immunocytochemistry, Western blot and RT-PCR analysis. However,the expres- sion of TGF-β1was higher in prostatic carcinoma cell line PC-3 than in prostatic epithelial cell line RWPE-1 (P〈0.05). Conclusion: The down-regulated expression of LKB1 ,while the excessive expression of TGF-β1 may play synergetic roles in the prostatic car- cinogenensis. LKB1 gene may be a potential target for researching the molecular of prostatic cancer tumorgenesis and evolution by ex-ploring the interactive mechanism of LKB1 and TGF-β1.
