摘  要:目的观察急性百草枯中毒大鼠肾组织中诱导型一氧化氮合酶(i NOS)的表达和细胞凋亡情况,探讨百草枯导致肾损伤发生的病理生理机制。方法将84只成年健康SD大鼠(雌雄各半)随机分为对照组、染毒组各42只,染毒组给予20%百草枯溶液25 mg/kg腹腔内一次性注射,对照组大鼠给予等量0.9%氯化钠溶液腹腔内注射。分别于注射1 d、3 d、7d、14 d、21 d、28 d、35 d,每组各取6只,在乙醚麻醉下,留取肾组织标本,免疫组织化学法检测iNOS蛋白表达水平,原位末端标记法(TUNEL法)标记凋亡细胞,RT-PCR检测肾组织中iNOS mRNA表达情况。结果染毒组不同时点大鼠肾组织中iNOS蛋白和iNOS mRNA表达量均明显高于对照组(P均<0.05),细胞凋亡率均明显高于对照组(P均<0.05)。结论急性百草枯中毒大鼠肾组织中iNOS含量和细胞凋亡率明显升高,它们可能参与了百草枯导致肾损伤的过程。Objective It is to observe the expression of inducible nitric oxide synthase (iNOS) and cell apoptosis in renal tissue in acute paraquat-intoxication rats, thus to explore the pathological and physiological mechanism of renal injury induced by paraquat. Methods Eighty-four healthy adult Sprague-Dawley rats (42 male, 42 female) were randomly divided into two groups:control group and poisoned group, each group had 42 rats. The poisoned group was given 20% paraquat liquid 25 mg/ kg once by intraperitoneal injection, while the control group was given the same dosage of normal saline by intraperitoneal in- jection. On lst,3rd,Tth,14th,21st,28th,and 35th day,six rats in each group were used to get their renal tissue to detect the levels of iNOS by immunohistochemistry, mark the apoptosis cells by TUNEL method, detect the expression of iNOS mRNA in renal tissue by RT - PCR method. Results Compared with control group, the levels of iNOS mRNA and protein in poisoned group were significantly higher while the apoptosis rate was significantly lower than that at every time point ( P 〈 0.05 ). Con- clusion The levels of inducible nitric oxide synthase (iNOS) and cell apoptosis in kidney of acute paraquat-induced rats were significantly increased. They could participate in the mechanism leading to renal damage.
