摘 要:目的探讨橙皮苷对IgA肾病大鼠肾脏组织中Nephrin表达是否有调节作用及其作用机制。方法选取雄性SD大鼠60只,适应性喂养2周后随机分为正常组、模型组和橙皮苷组,每组20只。除正常组外,其余组均造模。造模成功后,橙皮苷组给予橙皮苷10mg/(kg·d)灌胃,模型组和正常组给予生理盐水1 mg/(kg·d)灌胃,均1次/d。分别于造模6周、10周及灌胃干预4周、8周检测各组大鼠尿红细胞计数;分别于灌胃干预4周、8周检测24 h尿蛋白定量、血肌酐和尿素氮水平,采用免疫组化、Western blot法检测肾脏组织中Nephrin蛋白表达情况,采用RT-PCR法检测肾脏组织中Nephrin mRNA表达情况。结果与正常组相比,模型组大鼠毛发杂乱,饮食减少,橙皮苷组干预后毛色、饮食进水及活动度较模型组有一定改善。造模6周、10周及灌胃干预4周、8周,模型组和橙皮苷组尿红细胞计数均明显高于正常组(P均<0.05);灌胃干预4周、8周后,橙皮苷组尿红细胞计数均明显低于模型组(P均<0.05)。灌胃干预4周、8周后,模型组24 h尿蛋白定量、血肌酐和尿素氮水平均明显高于正常组(P均<0.05),Nephrin蛋白和mRNA表达均明显低于正常组(P均<0.05);灌胃干预4周后,橙皮苷组24 h尿蛋白定量和尿素氮水平均明显低于模型组(P均<0.05),Nephrin蛋白和mRNA表达均明显高于模型组(P均<0.05);灌胃干预8周后,橙皮苷组24 h尿蛋白定量、血肌酐和尿素氮水平均明显低于模型组(P均<0.05),Nephrin蛋白和mRNA表达均明显高于模型组(P均<0.05)。结论橙皮苷能明显降低IgA肾病大鼠24 h尿蛋白定量,减少尿红细胞计数,保护肾功能,这可能与其可上调肾脏组织中Nephrin蛋白和mRNA的表达有关。Objective It is to investigate whether hesperidin has regulation on Nephrin expression in kidney tissue in IgA nephropathy rats. Methods After adaptive feeding for 2 week, 60 male SD rats were randomly divided into normal group, model group and hesperidin treatment group, each group had 20 rats. Except normal group, model establishing was performed in the other groups. After successful model establishing, hesperidin treatment group was given hesperidin 10 mg/(kg· d) by lavage, while model group and normal group were given normal saline once per day. Urine red blood cell element was detected in 6, 10 weeks after model establishing and at 4 and 8 weeks' intervention in every group, and 24 h urine protein, serum cre- atinine and urine nitrogen content were determined at 4 and 8 weeks' intervention. Immunohistochemistry, Western blot and RT - PCR were used to detected the Nephrin mRNA and protein expression. Results Compared with normal group,the rats in model group was messy hair, their diet significantly reduced. After treatment, the rats' hair color, diet intake and activity were improved in hesperidin treatment group compared with model group. Compared with normal group, urine red blood cells increased significantly in model group and treatment group(P 〈 0.05) , but after intervention it was lower in treatment group than that in model group(P 〈0" 05). Compared with normal group, 24 h urinary protein, serum creatinine and urea nitrogen increased significantly in the other two groups. After 4 and 8 weeks' intervention, the levels of 24 h urinary protein and urea nitrogen were lower while the levels of Nephrin protein and mRNA expression were significantly higher in treatment group than that in model group( P 〈 0.05 ) ; After 8 weeks of treatment, the level of serum creatinine was also lower two ( P 〈 0.05 ). fur-ther increased.
