摘  要:采用水提醇沉法提取矮冷水花多糖,以多糖得率为指标,考察提取时间、提取温度、料液比及提取次数对多糖提取率的影响,在单因素试验基础上采用正交试验优化提取工艺参数。通过测定矮冷水花多糖清除·OH自由基能力,1,1-二苯基-2-苦基苯肼(1,1-diphenyl-2-picrylhydrazyl,DPPH)自由基及螯合力来评价其抗氧化活性。结果表明,矮冷水花多糖水提取的最佳工艺条件为提取温度100℃、提取时间4 h、料液比1∶40(g/m L)、提取次数4次,在该条件下,矮冷水花多糖的提取率为1.19%,以正交试验极差分析得出温度对矮冷水花粗多糖提取影响最大。矮冷水花多糖清除DPPH自由基、·OH自由基及金属螯合力均随多糖浓度的增加而上升;1.28 mg/m L的多糖对DPPH自由基的清除率达70.7%,对羟基自由基的清除率为41.1%,1.28 mg/m L多糖的金属螯合力在562 nm下吸光值为0.98。因此,矮冷水花多糖有较强的抗氧化能力。The polysaccharides of Pilea peploides (Gaudich.)Hook.et Am. was extracted by water extraction-alcohol precipitation method. The extraction process was optimized by investigating polysaccharide yield as the response value of extraction time, extraction temperature, solid to solvent ratio and extraction time. Based on the results of single-factor experiments ,orthogonal array experiments were carried out to determine the optimum process parameters. The antioxidant capacity of polysaccharides of Pilea peploides (Gaudich.)Hook.et Am. was evaluated by the scavenging activities against 1,1-diphenyl-2-picrylhydrazyl (DPPH), hydroxyl free radical (·OH )and chelating ability assays. The results showed that the best conditions for water extraction-alcohol precipitation were determined as follows :extraction temperature was 100 ℃; the time was 4 h ;solid to liquid ratio was 1:40 (g/mL) ;and the extraction time was 4 times,the extraction ration reached 1.19% .The scavenging capacity of .OH,DPPH and chelating ability increased with the increasing concentration of polysaccharide. 1.28 mg/mL concentration of polysaccharide had 70.7% scavenging ratio of DPPH ,and 41.1% scavenging ratio of· OH, absorbance at 562 nm of chelating ability was 0.98 when the concentration of polysaccharide was 1.28 mg/mL. From the results it could be concluded that polysaccharide from Pileapeploides (Gaudich.)Hook.et Am. had significant antioxidant activity in vitro.
