摘  要:目的:探索不同冷冻时间对造模的影响,为寒凝血瘀证动物模型的制作提供一种优化造模方法。方法:将48只大鼠按随机数字表法分为正常对照组、寒凝血瘀证3 d组、7 d组、14 d组、20 d组;瘀热互结证组6组各8只,寒凝组大鼠在-20℃持续受冻1 h左右,每日1次,持续时间分别为3、7、14、20 d;瘀热组大鼠采用角叉菜胶联合干酵母悬液造模。结果:与正常对照组比较,寒凝3 d组TSH、ADR显著增加,寒凝7 d组部分血流变学指标、ET显著增加,NO显著降低;寒凝14 d组血流变学指标、ET显著增加,NO、TSH、ADR显著降低;寒凝20 d组血流变指标、ET、NO趋于正常;TSH、ADR显著降低。结论:寒凝血瘀证大鼠最佳冷冻时间为14 d,这样既可保证血瘀证模型成功,同时也符合寒证所具有的机体功能活动衰减的特点。Objective: Show the effects of different freezing time and provide a new improving method for Blood Stasis due to Cold Syndrome( BSCS). Method: The forty-eight rats were divided randomly into 6 groups: normal control group,BSCS group for 3 d,7 d,14 d,20 d,Blood Stasis and Heat Accumulation Syndrome( BSHAS) group,8 in each. The rats of BSCS group were kept in- 20℃ ice 1h a day for 3,7,14,20 d. BSHAS group was made by using carrageen and dry yeast suspension. Results: Compared with the normal group,in 3 d of BSCS group,TSH,ADR improved significantly. In 7 d of BSCS group,part of hemorheology indexes and ET improved significantly; NO reduced obviously. In 14 d of BSCS group,hemorheology indexes and ET improved significantly; NO,TSH,ADR reduced obviously. In 20 d of BSCS group,TSH,ADR reduced obviously while hemorheology indexes,ET,NO became normal. Conclusion: The best freezing time for BSCS group is 14 d,which can not only ensure the success of the model of blood stasis syndrome,but also has the characteristics of the functional activity attenuation.
