摘  要:目的乳腺癌是女性最常见的恶性肿瘤之一,并且是女性癌症死亡的首要原因。本研究总结肝X受体(liver X receptor,LXR)及其激动剂的特征和它们在乳腺癌中的研究进展,探索LXR作用于乳腺癌的靶基因,为乳腺癌的靶向治疗寻找新靶点提供思路。方法通过查阅PubMed和CNKI数据库,以"肝X受体、乳腺癌、肿瘤、ABCA1和ABCG1"为关键词,检索2010-01-2016-01相关文献,共检索到英文文献249条,中文文献176条。纳入标准:肝X受体的特性;肝X受体在不同肿瘤中的机制研究;肝X受体与乳腺癌的相关性。排除标准:未涉及肝X受体及其靶基因的乳腺癌和其他肿瘤的研究。根据纳入标准,符合分析文献29篇,并加入原始文献3篇。结果 LXR激动剂对多种癌细胞均有抑制作用,但其内源性激动剂27HC在ER阳性乳腺癌中也可激活ER促进乳腺癌细胞增殖。LXR通过其靶基因三磷酸腺苷结合盒转运体A1(ATP binding cassette transporter A1,ABCA1)或三磷酸腺苷结合盒转运体G1(ATP binding cassette transporter G1,ABCG1)介导胆固醇代谢参与乳腺癌的发病机制。结论 LXR激动剂具有双向调节乳腺癌细胞的作用。LXR是通过ABCA1还是ABCG1参与乳腺癌的发病机制,目前尚存在争议。OBJECTIVE Breast cancer is one of the most common malignant tumors and the leading cause of cancer death among females. The features of liver X receptor (LXR) with its agonists and their research progress were reviewed in breast cancer, in order to explore the role of LXR target genes and provide new ideas to find new targets for targeted therapy of breast cancer. METHODS Based on PUBMED and CNKI databases, literatures were searched with the key words of "liver X receptor, breast cancer, tumour, ABCA1, ABCGI" from January 2010 to January 2016. Totally 249 English articles and 176 Chinese articles were got. Inclusion criteria: liver X receptor's characteristics; mechanism of liver X receptor in different tumors; correlation between liver X receptor and breast cancer. Rejection criteria: The study of breast cancer or other tumors did not involve the liver X receptor and its target genes. According to the retrieval criteria, 29 literatures and 3 original articles were finally analyzed. RESULTS LXR agonists can inhibit a variety of cancer cells, but 27HC as its endogenous agonist can also activate ER to promote the proliferation of breast cancer cells in ER positive breast cancer. LXR and its target genes ATP binding cassette transporter A1 (ABCA1) or ATP binding cassette trans- porter GI(ABCG1) are involved in the pathogenesis of breast cancer by mediating cholesterol metabolism. CONCLU- SIONS LXR agonists can regulate breast cancer cells in two directions. It is still a controversy that LXR participate in the pathogenesis of breast cancer through ABCA1 or ABCG1 and further studies are necessary.
