通过氨基酸渗漏检测小麦种子活力的研究

摘  要:本研究采用泰山22号和石麦20号两个小麦品种,依据含有氨基物质(氨基酸、蛋白质、多肽)与水合茚三酮共热产生蓝紫色化合物的原理,对茚三酮显色法是否适于小麦种子活力检测进行了研究。分别对小麦净种子进行人工加速老化处理0d、2d、4d、6d,以被处理小麦种子标准发芽试验得到的种子活力指标(发芽指数、活力指数)作为种子活力的标准。将种子每孔1粒置于96孔细胞培养板中25℃浸泡6h,吸取50μL种子渗出液于新96孔板中相应位置并加入100μL茚三酮反应试剂,80℃水浴反应30min,用酶标仪记录570nm吸光度值,与老化处理种子的标准发芽试验各项种子活力指标比较。结果表明两个品种小麦种子活力指标与茚三酮显色法测定的吸光度均呈极显著负相关,相关系数在-0.9837^-0.9988之间,将吸光度大于1.0的蓝色孔比例与种子活力指标进行相关分析,其相关系数在-0.9739^-0.9999之间,达到极显著,说明依据氨基酸渗漏的茚三酮显色法可以用于小麦种子活力的快速、非破坏检测。Two wheat varieties, Tai mountain 22 and Shi wheat 20 were used in this paper. The experiment is to explore the feasibility of seed vigor detection method of wheat with ninhydrin reaction, according to the principle of color reaction of nin- hydrin with amino- containing substances, including amino acids, proteins, and polypeptides, under the condition of heating to generated significant blue-violet compound of ninhydrin color. Pure seed materials were treated by artificial accelerated aging method 0d, 2d, 4d, 6d separately. Then the germination test was performed, with four replications per aging treatment, calculate seed vigor index, germination rate, germination index, vigor index after experiment. Wheat seeds were soaked individually in 80μL distilled water in 96-well microtiter plates for 6h at 25℃. Then 50μL of leaehate from each well was transferred to new plates and 100μL of ninhydrin was added to each well. The plates were then incubated for 30min at 80℃ for color devel0pmentl Recording color intensity and the absorbance values under 570nm, comparing with the seed vigor index from the standard germination test. The results show that the relationship between the average absorbance values of seed leachate measured at 570nm and seed vigor of two cultivars of wheat seeds have a very high negative correlation, with coefficient value ranging from -0. 9837 - -0. 9988. Similarly, the relationship between the proportion of the blue color wells (absorbance values higher than 1.0) and seed vigor of two cultivars of wheat seeds, high negative correlation, with co- efficient value ranging from -0. 9739- -0. 9999, which shows the ninhydrin reaction by detection amino acids leakage is quickly, nondestructive method for wheat seed vigor test.
