摘 要:目的:本研究旨在通过观察胃康宁对功能性消化不良大鼠胃排空功能及血浆胃肠激素水平的影响,探讨其对功能性消化不良的作用机制。方法:将雄性10日龄Wistar幼鼠50只,随机分为正常组15只、模型组15只、中药胃康宁组10只和阳性药物组10只,采用0.1%IA灌胃诱导大鼠FD模型。观察胃康宁对功能性消化不良大鼠食量、体重、胃排空功能等指标的影响,采用酶联免疫吸附测定法(ELISA)检测血浆MTL、Ghrelin、obestatin和血清GAS含量变化。结果:与正常组相比,模型组大鼠摄食量、体重和胃排空率降低(P<0.05),血浆MTL、Ghrelin和血清GAS含量降低,血浆obestatin水平升高;与模型组比较,胃康宁组和多潘立酮组小鼠食量、体重和胃排空率增加,血浆MTL、Ghrelin及血清GAS含量升高,血浆obestatin水平降低。结论:胃康宁对功能性消化不良模型大鼠具有改善胃肠动力、降低胃肠感觉过敏的作用,其治疗机制可能与调节外周血MTL、Ghrelin、obestatin及GAS含量的变化有关。Objective:To study the effect of weikangning on gastric emptying and plasma gastrointestinal hor‐mones in rats with functional dyspepsia (FD) ,and investigate its possible mechanism to regulate the Gastrointestinal func‐tion .Methods :The rats were randomly divided into fourgroups:model control group(n=15) ,domperidone group(n=10) ,Weikangning group(n=10) ,and normal control group(n=15);the change of intake of water and food ,body weight , and gastric emptying rate were observed .The levels of plasma motilin ,Ghrelin ,Obestatin and gastrin were measured by ELISA method .Results :Compared with the normal group ,the food intake ,body weight and gastric emptying rate were reduced (P<0 .05) ,the levels of plasma MTL ,Ghrelin and serum GAS were lower ,the levels of obestatin was higher in model group;While ,the food intake ,body weight and gastric emptying rate were increased ,plasma MTL ,Ghrelin and‐serum GAS were higher ,the level of plasma obestatin was lower in Weikangning group and domperidone group ,com‐pared with the model group .Conclusion:Weikangning can improve the gastric motility and reduce gastrointestinal hyper‐sensitivity in FD rats ,and its mechanism may be related to regulating the levels of plasma motilin ,Ghrelin ,Obestatin and gastrin .
