摘　　要：目的:研究p21激活激酶4(PAK4)在人乳腺癌细胞和组织中的表达和定位。方法:蛋白印迹法检测PAK4在人乳腺癌细胞系中蛋白和基因的表达,激光共聚焦扫描显微镜检测内源PAK4在MCF-7乳腺癌细胞中的定位。结果:PAK4在人乳腺肿瘤细胞系中蛋白和基因表达水平增高,激光共聚焦扫描显微镜观察发现PAK4主要定位于MCF-7细胞质中。结论:PAK4在人乳腺癌细胞系中高表达,主要定位于乳腺癌组织细胞质内,可能是人乳腺癌重要的调节因子。Objective：To investigate the expression and localization of p21 -activated kinase 4（PAK4）protein in human breast cancer.Methods：Immunoblot was used to analyze exogenous PAK4 expression from human breast cnac-er cell lines.The endogenous localization of PAK4 in MCF-7 cells was observed by using laser scanning confocal mi-croscopy.Results：PAK4 mRNA and protein expression was high in human breast cancer cells.The human PAK4 pro-tein was mainly localized in the cytoplasm by laser scanning confocal microscopy.Conclusion：PAK4 was highly ex-pressed in human breast cancer cell lines and localized mainly in the cytoplasm,which may be an important regulator in human breast cancer.
